IL-17、RORγt在百草枯中毒小鼠急性肺损伤中的表达

【摘要】:目的:观察Th17型细胞因子IL-17A和特异性转录因子维甲酸相关孤独受体(RORγt)在百草枯(PQ)中毒小鼠急性肺损伤的表达及意义。方法:雄性昆明小鼠随机分为对照组和模型组,各8只。模型组予一次性腹腔注射20 mg/kg PQ建立染毒模型,对照组予同法注射等量生理盐水,48 h后处死小鼠并取材。HE染色观察肺组织病理变化;原位末端缺刻标记法观察细胞凋亡并计数;酶联免疫吸附法检测血清IL-17A的水平;免疫荧光定量PCR法和蛋白质免疫印迹法检测肺组织RORγt m RNA及IL-17蛋白的表达。结果:与对照组相比,模型组小鼠肺组织炎症及细胞凋亡显著增加;血清IL-17A含量明显增多(P0.05);肺组织RORγt m RNA和IL-17蛋白的表达均增高。结论:Th17可能通过分泌IL-17参与PQ中毒小鼠急性肺损伤的炎症过程,并在其致病中发挥重要作用。
[Abstract]:Aim: to investigate the expression and significance of Th17 type cytokine IL-17A and specific transcription factor retinoic acid-associated lonely receptor (ROR 纬 t) in acute lung injury induced by paraquat (PQ) in mice. Methods: male Kunming mice were randomly divided into control group and model group with 8 mice each. The model group was injected intraperitoneally for 20 mg/kg PQ and the control group was injected with the same amount of normal saline. After 48 hours, the mice were killed and the pathological changes of lung tissue were observed by HE staining. Apoptosis and count were observed by in situ terminal dehiscence labeling, serum IL-17A levels were detected by enzyme-linked immunosorbent assay (Elisa), and the expression of ROR 纬 t m RNA and IL-17 protein in lung tissue were detected by immunofluorescence quantitative PCR and Western blot. Results: compared with the control group, the lung tissue inflammation and apoptosis in the model group were significantly increased, the serum IL-17A content was significantly increased (P0.05), and the expression of ROR 纬 t m RNA and IL-17 protein were increased in the lung tissue of the model group. Conclusion: Th17 may participate in the inflammatory process of acute lung injury in mice poisoned by PQ by secreting IL-17 and play an important role in its pathogenesis.
