白细胞介素-6基因多态性与脓毒症的相关性研究

【摘要】:目的:研究白细胞介素-6基因启动子区rs1800796C/G的等位基因及其基因型在广西地区人群中的分布频率,探讨IL-6rs1800796C/G基因多态性与脓毒症的关系。 方法:采用单碱基延伸的PCR技术和DNA测序法,检测199例广西健康体检人群和152例脓毒症患者的IL-6基因启动子区rs1800796C/G多态性,并分析其多态性与脓毒症的相关性。 结果:广西人群IL-6基因启动子区rs1800796位点存在G、C两种等位基因及CC、CG、GG三种单核苷酸多态基因型。其基因型及等位基因频率在男女组间比较差异无统计学意义(P>0.05),两种等位基因的分布在脓毒症患者与对照组间无显著差异(P>0.05)。与非洲人、欧洲人、日本人和北京人等四个人群的SNP分型数据进行比较,广西人群的基因型及等位基因分布频率与日本人和北京人比较差异无统计学意义(均P>0.05),而与欧洲人、非洲人比较差异有统计学意义(均P<0.05)。广西人、北京人和日本人CC基因型所占比例较高,,而非洲人和欧洲人GG型频率较高。脓毒症患者血清IL-6水平明显增高(P<0.05),IL-6rs1800796C/G基因多态性与脓毒症患者血清IL-6水平无相关性。 结论:广西人群的IL-6基因启动子区rs1800796C/G存在基因多态性,其基因型分布及等位基因分布频率与性别无相关性,但与其他种族人群相比存在明显差异。IL-6rs1800796C/G基因多态性与脓毒症易感性和血清IL-6水平无相关性。
[Abstract]:Objective: to study the alleles of interleukin-6 (IL-6) gene promoter region rs1800796C/G and its genotype distribution in Guangxi population, and to explore the relationship between IL-6rs1800796C/G gene polymorphism and sepsis. Methods: single base extension PCR technique and DNA sequencing were used to detect the rs1800796C/G polymorphism of IL-6 gene promoter in 199 healthy controls and 152 sepsis patients, and the correlation between IL-6 gene promoter polymorphism and sepsis was analyzed. Results: there were two alleles in rs1800796 locus of IL-6 gene promoter and three single nucleotide polymorphism genotypes of CC,CG,GG in Guangxi population. There was no significant difference in genotype and allele frequency between male and female groups (P > 0.05), but there was no significant difference in the distribution of the two alleles between septic patients and controls (P > 0.05). Compared with the SNP genotyping data of African, European, Japanese and Beijingese, there was no significant difference in genotype and allele distribution between Guangxi population and Japanese population and Beijingese population (all P > 0. 05). The difference between African and European was statistically significant (all P < 0.05). The proportion of CC genotypes was higher in Guangxi, Beijing and Japan, but the frequency of GG was higher in Africans and Europeans. Serum IL-6 level was significantly higher in septic patients (P < 0. 05). There was no correlation between IL-6rs1800796C/G gene polymorphism and serum IL-6 level in septic patients. Conclusion: there is a polymorphism of rs1800796C/G in the promoter region of IL-6 gene in Guangxi population, and there is no correlation between genotype distribution and allele distribution. IL-6rs1800796C/G gene polymorphism was not associated with susceptibility to sepsis and serum IL-6 level.
