辛伐他汀对脓毒症早期大鼠血脂代谢及肝脏氧化应激的影响

【摘要】:目的探讨辛伐他汀对脂多糖(LPS)致大鼠脓毒症模型早期血脂代谢及肝脏氧化应激的影响。方法将56只SD雄性大鼠按随机数字表法分成对照组(8只)、模型组(24只)、辛伐他汀组(24只)。腹腔注射LPS 10 mg/kg复制脓毒症大鼠模型。对照组和模型组于腹腔注射生理盐水或LPS前给予生理盐水1 mL/d灌胃,辛伐他汀组于制模前给予辛伐他汀20 mg/(kg·d)灌胃,均连续1周。于制模后6、12、24 h测定大鼠血清总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)水平和肝脏组织超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量。结果与对照组比较,模型组大鼠血清TC(mmol/L)、TG(mmol/L)、LDL(mmol/L)水平和肝脏组织MDA(nmol/mg)含量呈先升高后降低表现,而肝脏组织SOD活性均明显降低(P0.05)。与模型组比较,辛伐他汀组6 h血清TG水平和24 h血清TC水平显著降低(P0.05),6 h、12 h肝脏组织MDA含量显著降低(P0.05),辛伐他丁组和模型组比较,各时点肝脏SOD活性差异无统计学意义(P0.05)。结论辛伐他汀降低了脓毒症早期大鼠血清总胆固醇水平,且能减轻肝脏的氧化应激损伤。
[Abstract]:Objective to investigate the effects of simvastatin on early lipid metabolism and oxidative stress in liver of rats with sepsis induced by lipopolysaccharide (LPS). Methods 56 male SD rats were randomly divided into control group (n = 8), model group (n = 24) and simvastatin group (n = 24). The rat model of sepsis was established by intraabdominal injection of LPS 10 mg/kg. The control group and the model group were given saline 1 mL/d before intraabdominal injection of saline or LPS, and simvastatin 20 mg/ (kg 路d) was given intragastrically for one week. The level of serum total cholesterol (TC), triacylglycerol (TG), (HDL), low density lipoprotein (LDL), the activity of SOD (SOD) and the content of malondialdehyde (MDA) in liver tissue were measured at 6, 12 and 24 h after modeling. Results compared with the control group, the level of serum TC (mmol/L), TG (mmol/L), LDL (mmol/L) and the content of MDA (nmol/mg) in liver tissue of the model group increased at first and then decreased, while the SOD activity of liver tissue decreased significantly (P 0.05). Compared with the model group, the serum TG level and 24 h serum TC level in simvastatin group decreased significantly at 6 h and 24 h (P 0.05), and the MDA content in liver tissue decreased significantly at 12 h (P 0.05). There was no significant difference in SOD activity between simvastatin group and model group at each time point (P 0.05). Conclusion Simastatin can reduce the level of serum total cholesterol in early sepsis rats and reduce the oxidative stress injury of liver.
