重症急性胰腺炎大鼠血清Apelin、C反应蛋白及肿瘤坏死因子-α水平变化及临床意义

【摘要】:目的探讨重症急性胰腺炎(SAP)大鼠血清Apelin、C反应蛋白(CRP)及肿瘤坏死因子(TNF)-α水平的变化及临床意义。方法将36只SD雄性大鼠按照随机数字表法分为SAP组、假性手术组、对照组,每组12只,SAP组按照大鼠体重1 ml/kg剂量胆总管注入50 g/L牛磺胆酸钠制备SAP模型,假性手术组行SAP组相同手术但不注射药物,对照组正常饲养。采用酶联免疫吸附实验(ELISA)法测定Apelin、CRP及TNF-α水平,光镜观察SAP胰腺组织变化并进行组织病理评分。结果 SAP组胰腺组织严重水肿,组织细胞坏死、具有片状出血斑并且胰腺出现局部皂化斑,假性手术组正常或者轻微水肿,无明显异常,对照组组织正常,建模成功。SAP组血清CRP和TNF-α水平均高于假性手术组和对照组,Apelin明显低于假性手术组和对照组(P0.05)。SAP组胰腺组织病理学评分高于假性手术组和对照组(P0.05)。SAP组血清Apelin水平与胰腺组织病理学评分呈负相关(r=-0.876,P0.05),CRP和TNF-α水平与胰腺组织病理学评分呈正相关(r=0.698、0.724,P0.05)。结论血清Apelin、CRP及TNF-α水平变化与SAP发生、发展密切相关,可以作为疾病严重程度判断和治疗效果评价的重要指标。
[Abstract]:Objective to investigate the changes and clinical significance of serum Apelin,C reactive protein (CRP) and tumor necrosis factor (TNF)-伪 in rats with severe acute pancreatitis (SAP). Methods 36 male SD rats were randomly divided into SAP group (n = 12), control group (n = 12) and control group (n = 12). SAP model was established by injecting 50 g / L sodium taurocholate into common bile duct at a dose of 1 ml/kg in SAP group. SAP group was treated with the same operation but not injected with drugs, and the control group was fed normally. The levels of Apelin,CRP and TNF- 伪 were measured by enzyme-linked immunosorbent assay (ELISA). The changes of pancreatic tissue in SAP were observed by light microscope and the pathological score was evaluated. Results the pancreatic tissue of SAP group was severe edema, tissue cell necrosis, flake bleeding spot and local saponification spot of pancreas. There was no obvious abnormality in false operation group, and the control group was normal, and the model was successful. The levels of serum CRP and TNF- 伪 in SAP group were higher than those in pseudo operation group and control group.
