【摘要】:目的: 通过慢性不可预见刺激(CUS)致大鼠抑郁行为探讨性别差异与神经细胞单胺氧化酶B(MAO-B)、5-羟色胺转运体(5-HTT)及去甲肾上腺素转运体(NET)表达的关系。结合临床具有自杀行为的抑郁患者,探讨其自杀行为是否存在性别差异。 方法: 1. SD大鼠雌雄两性各30只,随机分为雄对照组(MCG)、雄模型组(MMG)、雌对照组(FCG)和雌模型组(FMG),采用孤养结合CUS方式建立大鼠抑郁模型。 2.开场实验和强迫游泳实验评价大鼠焦虑抑郁行为。 3. HE染色镜下观察大鼠海马病理形态改变。 4.Western blot(WB)法和荧光定量PCR法分别测定脑桥NET和海马5-HTT、MAO-B蛋白及mRNA表达。 5.计算机检索中国知网学术期刊数据库、Pubmed等中英文数据库,获得抑郁症致男女自杀行为的相关文献。依照一定标准进行资料筛选并将合格资料进行Meta分析。 结果: 1.经CUS处理的FMG和MMG大鼠开场实验水平得分和垂直得分显著低于各自对照组,,且MMG大鼠水平得分、垂直得分显著低于FMG;FMG和MMG大鼠强迫游泳不动时间显著长于各自对照组,且MMG大鼠不动时间显著长于FMG。 2.MMG、FMG组大鼠海马细胞有明显的核固缩、深染等损伤现象,FMG组损伤程度似不如MMG组严重。 3.与各自对照组相比,FMG、MMG大鼠5-HTT表达均下降,NET和MAO-B表达均升高;且MMG大鼠脑桥NET表达显著高于FMG大鼠。 4.临床资料的Meta分析结果显示,男性与女性总自杀行为比值比(odds ratio,OR)为0.84(95%CI:0.63—1.12,P=0.23)。自杀死亡的OR值为0.94(95%CI:0.18—5.07,P=0.94),自杀未遂OR值为0.75(95%CI:0.59—0.95,P=0.02)。 结论: CUS致大鼠抑郁行为存在显著的性别差异,此差异可能与影响单胺递质代谢的MAO-B、NET、5-HTT表达量的变化不同有关。男女抑郁症患者自杀行为存在一定性别差异,男性自杀未遂的发生率显著高于女性。
[Abstract]:Objective: To explore the relationship between sex difference and neuronal monoamine oxidase (BMAO-B-) in depression induced by chronic unpredictable stimulation (CUS) in rats. The relationship between serotonin transporter (5-HTT) and norepinephrine transporter (NET). Methods: 1. Thirty male and female SD rats were randomly divided into male control group (MCG), male model group (MMGN), female control group (FCGG) and female model group (FMG). The depression model of rats was established by using solitary nourishment combined with CUS. 2. The behavior of anxiety and depression in rats was evaluated by open field experiment and forced swimming experiment. 3. The histopathological changes of hippocampus were observed by HE staining. 4. The expression of MAO-B protein and mRNA in pons NET and hippocampus were detected by Western blottle and fluorescence quantitative PCR, respectively. 5. Computer retrieval of Chinese and English databases such as online academic journals. To obtain the relevant literature on suicidal behavior of men and women with depression. To screen the data according to certain criteria and to analyze the qualified data by Meta. Results: 1. The open field test level and vertical score of FMG and MMG rats treated with CUS were significantly lower than those of the control group, and the horizontal score and vertical score of MMG rats were significantly lower than those of the control group. The immobility time of FMG and MMG rats was significantly longer than that of their control groups, and the immobility time of MMG rats was significantly longer than that of FMGs. 2. The damage degree of hippocampal cells in MMG- FMG group was not as serious as that in MMG group. 3.Compared with the control group, the expression of 5-HTT decreased and the expression of net and MAO-B increased in MMG rats. The expression of NET in MMG rats was significantly higher than that in FMG rats. 4. The Meta analysis of clinical data showed that the ratio of total suicide behavior between male and female was 0.8495 CI: 0.63-1.12. The OR value of suicide death was 0.94% 95% CI: 0.18-5.07% P0.94). The OR value of attempted suicide was 0.75% 95% CI: 0.59-0.95% P0.02. Conclusion: There were significant gender differences in depressive behaviors induced by CUS in rats, which may be related to MAO-BN, which affects the metabolism of monoamine transmitters. The changes of 5-HTT expression were related to different levels of 5-HTT expression. There were some gender differences in suicidal behavior between male and female patients with depression, and the incidence of attempted suicide in male was significantly higher than that in female.
