白藜芦醇对脂多糖诱导PC12细胞炎症的保护作用的研究

【摘要】:目的:通过构建脂多糖(Lipopolysaccharide, LPS)诱导的大鼠肾上腺嗜铬细胞瘤细胞株PC12细胞炎症模型,建立阿尔茨海默病的体外模型;从细胞、分子层面探讨白藜芦醇(Resveratrol,RES)对PC12细胞炎症的保护作用。方法:体外培养的PC12细胞随机分成空白对照组、LPS (1mg/ml,18h)处理组、不同浓度的白藜芦醇(25、50和100μM,1h)+LPS处理组。采用免疫荧光双标记检测方法(Double-immunofluorescence Labeling)在荧光显微镜下观察细胞内白介素-1β(interleukin-1β, IL-1β)和肿瘤坏死因子-α(tumor necrosis factor-α, TNF-α)的表达变化;应用逆转录聚合酶链式反应(reverse transcription polymerase chain reaction, RT-PCR)在mRNA水平检测TNF-α和IL-1p的表达变化。 结果:不同浓度的白藜芦醇(25、50和100μM)在翻译和转录水平明显抑制了LPS诱导的PC12细胞炎性细胞因子IL-1β和TNF-α的表达,并呈现浓度依赖性抑制。 结论:白藜芦醇能够抑制脂多糖诱导的PC12细胞炎性细胞因子IL-1β和TNF-α的释放,从而发挥其抗神经炎症的作用。
[Abstract]:Objective: to establish an in vitro model of Alzheimer's disease in rat adrenal pheochromocytoma cell line PC12 cells induced by lipopolysaccharide (LPS). To investigate the protective effect of resveratrol resveratrolon resveratrol on inflammation of PC12 cells at the molecular level. Methods: PC12 cells cultured in vitro were randomly divided into two groups: the control group treated with 1 mg / ml of resveratrol for 18 h. The expression of interleukin-1 尾 interleukin-1 尾 interleukin-1 尾 (IL-1 尾) and tumor necrosis factor- 伪 (TNF- 伪) were observed under fluorescence microscope in different concentration groups treated with resveratrol 2550 and 100 渭 m for 1 h) LPS. Reverse transcription polymerase chain reaction (RT-PCRR) was used to detect the expression of TNF- 伪 and IL-1p at mRNA level. Results: different concentrations of resveratrol 2550 and 100 渭 M inhibited the expression of PC12 inflammatory cytokines IL-1 尾 and TNF- 伪 in a dose-dependent manner at the translation and transcription levels. Conclusion: resveratrol can inhibit the release of inflammatory cytokines IL-1 尾 and TNF- 伪 in PC12 cells induced by lipopolysaccharide, thus exerting its anti-neuritis effect.
