应用点着色聚类方法识别阿尔茨海默病致病基因

【摘要】:阿尔茨海默病是一种最常见的痴呆症。识别与阿尔茨海默病有关的候选致病基因是一项重要的研究。目前通过分析DNA微阵列数据从而识别与疾病有关的基因已经成为一个热点,并且在这些分析方法中,聚类是计算分析的一个基础。在本文中,应用图论中的点着色理论,将表达相似的基因聚在一起(即生物功能相近的基因聚在一起)。然后分析聚类后的结果,识别出可能与阿尔茨海默病有关的基因。本文提出了在图论的点着色基础上一种全新的聚类识别方法。运用这种方法,识别了27个基因与阿尔茨海默病有关,并且其中3个基因已经被其他研究小组证明与阿尔茨海默病有关,另外2个基因与神经疾病有关。本文的方法有如下三个优点:1.能将共同表达的基因聚在一起;2.聚类后得到类的个数在理论上是最少的;3.使用点着色方法对基因进行聚类和识别在过去的研究中从没被提出过。本文主要研究内容如下: (1).介绍了阿尔茨海默病的研究现状和DNA微阵列技术,并介绍DNA微阵列技术在阿尔茨海默病上的应用。 (2).详细介绍了图论中点着色理论和聚类的关系,同时阐述了点着色聚类算法。 (3).应用点着色算法对基因进行聚类,识别与阿尔茨海默病相关的基因,最后对比点着色聚类算法与K-means算法
[Abstract]:Alzheimer's disease is the most common form of dementia. The identification of candidate genes associated with Alzheimer's disease is an important study. At present, the identification of disease-related genes by analyzing DNA microarray data has become a hot spot, and clustering is the basis of computational analysis in these analytical methods. In this paper, the point coloring theory in graph theory is applied to gather genes expressing similar functions (that is, genes with similar biological functions). The clustering results were then analyzed to identify genes that might be associated with Alzheimer's disease. In this paper, a new clustering recognition method based on point coloring of graph theory is proposed. Using this method, 27 genes were identified to be associated with Alzheimer's disease, and three of them have been linked to Alzheimer's disease by other research teams, and the other two genes have been linked to neurological diseases. The method in this paper has three advantages: 1. A gene that can be co-expressed together. After clustering, the number of classes is the least in theory. The use of point-coloring to cluster and recognize genes has never been proposed in previous studies. The main contents of this paper are as follows: There is no such thing as a bird. This paper introduces the research status and DNA microarray technology of Alzheimer's disease, and introduces the application of DNA microarray technology in Alzheimer's disease. There are two pieces of water. The relationship between point coloring theory and clustering in graph theory is introduced in detail. 3. The point coloring algorithm is used to cluster genes and identify the genes related to Alzheimer's disease. Finally, the point coloring clustering algorithm and the K-means algorithm are compared.
