精神分裂症患者血清差异microRNA谱的检测

【摘要】:目的目前精神分裂症的临床诊断缺乏统一的标准。文中研究精神分裂症患者血清中差异miRNA表达谱,为该病的临床诊断提供依据。方法采用Flash TagTMBiotin RNA芯片技术和SAM软件筛选精神分裂症患者与健康受试者血清中差异表达的miRNA,用实时荧光定量逆转录PCR(RT-PCR)进行验证。结果筛选出3个差异表达的miRNA,其中hsa-miR-1281上调,表达倍数变化为1.508 81;hsa-miR-2861、hsa-miR-638下调,表达倍数变化分别为0.641 93和0.516 24。实时定量PCR结果证实,精神分裂症患者较健康受试者hsa-miR-2861与hsa-miR-638表达下调,分别为[(0.037±0.037)vs(3.159±2.761),χ2=4.089,P0.05]及[(0.006±0.006)vs(0.689±0.314),χ2=4.083,P0.05],hsa-miR-1281在精神分裂症患者组呈高表达[(1.221±0.041)vs(0.145±0.036),χ2=5.333,P0.05)。结论精神分裂症患者血清中存在差异表达的miRNA谱,miR-1281、miR-2861和miR-638可作为诊断精神分裂症的一个新的血清标志物。
[Abstract]:Objective at present, the clinical diagnosis of schizophrenia is lack of uniform standard. The differential miRNA expression profile in the serum of schizophrenic patients was studied in order to provide evidence for clinical diagnosis of schizophrenia. Methods Flash TagTMBiotin RNA microarray and SAM software were used to screen the differentially expressed miRNAs in the serum of schizophrenic patients and healthy subjects. Results three differentially expressed miRNAs were screened out, in which hsa-miR-1281 was up-regulated, and the expression multiple was down-regulated by 1.508 81hsa-miR-2861hsa-miR-638, and the expression multiple was 0.641 93 and 0.516 2424 respectively. The results of real-time quantitative PCR showed that the expression of hsa-miR-2861 and hsa-miR-638 were down-regulated in schizophrenic patients (0.037 卤0.037)vs(3.159 卤2.761g, 蠂 2 + 4.089U, P 0.05) and [0.006 卤0.006)vs(0.689 卤0.314g, 蠂 2 + 4.083P 0.05] hsa-miR-1281 were highly expressed in schizophrenic patients [1.221 卤0.041)vs(0.145 卤0.036, 蠂 2 5.333P 0.05, respectively]. Conclusion the differential expression of miR-1281miR-2861 and miR-638 in the serum of schizophrenic patients may be a new serum marker for the diagnosis of schizophrenia.
