血浆聚集素及其基因多态性与阿尔茨海默病的关系

【摘要】:目的:探讨中国汉族人聚集素在阿尔茨海默中的含量和意义以及阿尔茨海默病(AD)与聚集素基因的多态性的关系。 方法: 1.收集AD组与对照组的病例。 2.两组患者均取3ml外周静脉血。 3.采用低渗溶血、常规酚-氯仿法提取白细胞基因组中的DNA。 4.应用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)的技术观察聚集素基因多态性。 5.采用酶联免疫吸附测定法(ELISA)检测AD患者(AD组)与健康老年人(对照组)血浆聚集素水平。 结果: 1.AD组聚集素基因C/C基因型、等位基因C分布明显高于对照组(χ2=6.673,P0.05)(χ2=15.060,,P0.05);聚集素等位基因C、基因型C/C与AD呈显著正相关(OR=3.685,95%Cl=1.855-7.320)(OR=2.750,95%Cl=1.260-6.002)。 2.AD组血浆聚集素水平显著高于对照组,差异具有统计学意义(P0.01)。 结论: 1.聚集素基因C/C基因型、等位基因C与阿尔茨海默病相关联,且可能是AD的一种危险因素。 2.AD患者血浆聚集素含量较正常对照组明显升高,该结论与国外的研究报道一致。聚集素可能在AD的发生、发展中起着重要作用。
[Abstract]:Objective: to investigate the content and significance of agglutinin in Alzheimer's disease (AD) and its association with polymorphism of agglutinin gene in Chinese Han nationality. Methods: 1. The cases of AD group and control group were collected. 2. The peripheral venous blood of 3ml was taken from both groups. DNA.4 was extracted from leukocyte genome by hypotonic hemolysis and routine phenol-chloroform method. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique was used to observe the polymorphism of aggregin gene. The levels of plasma aggregates in AD patients (AD group) and the healthy elderly (control group) were detected by Elisa. Results: 1. The C / C genotype of the agglutinin gene in AD group was determined by enzyme linked immunosorbent assay (Elisa). The distribution of allele C was significantly higher than that of the control group (蠂 26.673, P 0.05) (蠂 2 = 15.060, P 0.05), and the genotype C / C and C / C were significantly positively correlated with AD. Conclusion: 1.855-7.320cl 1.855-7.320Cl1.75095Cl1.260-6.0020.2.The level of plasma aggregation in AD group was significantly higher than that in control group (P 0.01). C / C genotype and allele C are associated with Alzheimer's disease and may be a risk factor for AD. 2. Plasma concentration of aggregin in AD patients is significantly higher than that in normal controls. Agglutinin may play an important role in the occurrence and development of AD.
