不同浓度β-淀粉样蛋白对大鼠小胶质细胞特异性蛋白CD11b表达的影响

【摘要】:目的探讨免疫细胞膜表面整合素(CD11b)在β-淀粉样蛋白(Aβ)所引起阿尔茨海默病(AD)中的作用、机制及与Aβ剂量的关系。方法雄性Wistar大鼠50只,随机分为A、B、C、D、E 5组,每组10只;A组大鼠双侧海马CA1区注射5μl生理盐水,B~E组双侧海马注射5μl Aβ(分别含Aβ_(25~35)0.5、1、5、10μg);HE染色观察五组大鼠海马CA1区细胞形态及数量变化;免疫组化(IHC)检测海马CA1区Aβ、CD11b表达;实时荧光定量PCR技术(qRT-PCR)检测CD11b mRNA表达。结果 HE染色,光镜下观察D组、E组大鼠海马CA1区神经细胞数明显少于A组、B组、C组(P0.01);IHC检测D组、E组两组Aβ阳性表达率均高于A组、B组、C组(P0.01);D组、E组CD11b阳性表达率明显高于A组、B组、C组(P0.01);qRT-PCR检测大鼠海马组织内CD11b mRNA表达D组、E组组明显高于A组、B组、C组(P0.01)。结论 Aβ可激活小胶质细胞,募集MG到Aβ沉积部位,Aβ沉积量和CD11b表达量呈正相关。
[Abstract]:Objective to investigate the role and mechanism of immunomembrane integrin (CD11b) in (AD) of Alzheimer's disease induced by 尾 -amyloid protein (A 尾) and its relationship with A 尾 dose. Methods Fifty male Wistar rats were randomly divided into two groups: group A (n = 10) and group A (n = 10). In each group, 5 渭 l A 尾 (containing A 尾 _ (25) 35) 0.5A ~ (1 +) 510 渭 g was injected into the bilateral hippocampal CA1 area of rats in each group, and the morphological and quantitative changes of CA1 cells in the hippocampus of the five groups were observed by HE staining. Immunohistochemical (IHC) was used to detect the expression of A 尾 -CD11b in hippocampal CA1 and real-time quantitative PCR (qRT-PCR) was used to detect the expression of CD11b mRNA. Results HE staining, The number of CA1 neurons in the hippocampus of group D was significantly less than that of group A (P0.01). The positive expression rate of A 尾 in group D was higher than that in group C (P 0.01). The positive rate of CD11b in group E was significantly higher than that in group A (P 0.01). The expression of CD11b mRNA in hippocampus of group D was significantly higher than that of group C (P0.01). Conclusion A 尾 can activate microglia cells, and there is a positive correlation between the amount of A 尾 deposition and the expression of CD11b in recruiting MG to A 尾 deposition site.
