硫化氢对创伤后应激障碍大鼠空间学习记忆能力及海马神经元放电的影响
[Abstract]:Objective to investigate the behavioral changes and the changes of cysta-thionine- 尾 -synthase (cysta-thionine- 尾-synthase,CBS) / hydrogen sulfide (hydrogen sulfide,H_2S) levels in the brain of rats with post-traumatic stress disorder (posttraumatic stress disorder,PTSD). The effect and mechanism of exogenous H 2S supplementation on behavior of rats were studied. Methods Morris water maze was used to detect the spatial learning and memory ability of rats, and methylene blue method was used to detect the content of CBS/H_2S system in hippocampus. The discharge of hippocampal neurons was recorded by multitube microelectrode in external somatic electrophysiology. Results (1) compared with the normal control group, the escape latency of PTSD rats increased (P0.01), the percentage of staying time in the quadrant of the platform and the number of times passing through the original platform decreased significantly (P0.01). Compared with PTSD group, the content of CBS/H_2S system in hippocampus decreased significantly (P0.01). (2), and the escape latency of PTSD group (sodium hydrosulfide,NaH S) decreased (P0.01). The percentage of residence time in the quadrant of the platform and the number of times through the original platform position increased significantly (P0.01). (3). Microinjection of L-cysteine (L-cysteine) significantly increased the spontaneous discharge frequency of hippocampal neurons. (P0. 01). Conclusion the ability of spatial learning and memory in PTSD rats is reduced by inhibiting the CBS/H_2S system in hippocampal area. The mechanism of H2S improving spatial learning and memory ability of PTSD rats may be related to the increase of firing frequency and excitability of hippocampal neurons. The ability of spatial learning and memory was decreased by CBS/H_2S system in hippocampal area. The mechanism of H2S improving spatial learning and memory ability of PTSD rats may be related to the increase of firing frequency and excitability of hippocampal neurons.
