创伤后应激障碍大鼠海马、杏仁核神经元自噬和凋亡改变!

【摘要】:目的观察创伤后应激障碍(PTSD)模型大鼠海马、杏仁核神经元自噬和凋亡的改变,探讨PTSD海马、杏仁核体积异常的机制。方法成年健康雄性SD大鼠40只,随机分为对照组和模型组。采用改良的单一连续应激方法制备PTSD大鼠模型;采用Western blotting方法检测海马、杏仁核组织LC-3和Beclin-1的表达水平;TUNEL染色和流式细胞术检测海马、杏仁核神经元凋亡情况。结果模型组海马、杏仁核组织LC3-Ⅱ/LC3-Ⅰ比值和Beclin-1表达水平高于对照组(P0.05);模型组海马、杏仁核神经元TUNEL阳性细胞百分率和凋亡率高于对照组(P0.01)。结论 PTSD模型大鼠海马、杏仁核神经元存在明显的细胞自噬和凋亡,这可能是海马、杏仁核体积异常的机制之一。
[Abstract]:Objective to observe the changes of autophagy and apoptosis of hippocampal and amygdaloid neurons in rats with post-traumatic stress disorder (PTSD) and to explore the mechanism of abnormal volume of hippocampus and amygdala in PTSD. Methods 40 healthy male SD rats were randomly divided into control group and model group. The PTSD rat model was established by modified single continuous stress method. The expression of LC-3 and Beclin-1 in hippocampus and amygdala was detected by Western blotting method.
