犬尿氨酸通路相关物质在大鼠慢性脑低灌注致认知障碍中的作用
[Abstract]:Objective to investigate the expression of interferon- 纬 (IFN- 纬), TNF- 伪, inducible NO synthase (iNOS),) indole 3-dioxygenase (IDO) and canine uremic acid (KYN) in hippocampus of rats with chronic cerebral hypoperfusion. To investigate the changes of KYN pathway related metabolites in hippocampus of rats with cognitive impairment induced by chronic cerebral hypoperfusion. Methods Thirty Wistar rats were randomly divided into sham-operated group (n = 15) and bilateral common carotid artery permanent ligation group (2VO group) (n = 15). Morris water maze was used to detect the changes of spatial learning and memory in rats with chronic cerebral hypoperfusion. The expression of iNOS, interferon 纬 and TNF- 伪 in CA1 region of hippocampus was detected by immunohistochemistry, and the levels of IDO and KYN were detected by ELISA method. Results compared with the sham operation group, the escape latency of 2VO group was significantly prolonged, the percentage of swimming distance in the platform quadrant was significantly decreased, and the positive expression of iNOS, interferon 纬 and TNF- 伪 were observed in the 2VO group on the 3rd day. The levels of IDO and KYN were significantly increased [(4.73 卤0.21) ng/ml vs (0.89 卤0.06) ng/ml and (1.55 卤0.12) ng/ml vs (0.19 卤0.02) ng/ml,P0.05]. Conclusion the pathophysiological process of chronic cerebral perfusion rats includes inflammatory response, which may stimulate the high expression of IDO and KYN, and the accumulation of KYN and IDO may participate in the process of cognitive impairment.
