吗啡调节大鼠催产素合成与释放的机制及GIRK在吗啡效应中作用的研究

长期应用阿片可引起机体对阿片的依赖和耐受,但其机制目前尚不清楚, 这是神经科学工作者面临的重要研究课题之一。研究表明,下丘脑合成的神经 垂体激素催产素(OT)参与药物成瘾和学习记忆过程,减弱外周和中枢注射吗 啡引起的依赖和耐受,并减轻大鼠对海洛因的耐受和海洛因的自身给药行为. 侧脑室或边缘前脑内注射OT受体拮抗剂可易化吗啡的耐受,提示中枢OT可 拮抗吗啡的依赖和耐受的形成。急性吗啡处理可抑制大鼠下丘脑视上核(SON) 神经元的放电,并明显抑制OT的释放;慢性吗啡处理,可导致中枢OT含量 降低,戒断期间血浆OT水平出现瞬间的大量增加,表明中枢OT含量的变化 与吗啡依赖的形成有密切关系。阿片抑制视上核OT的释放,减轻OT对阿片 依赖和耐受的拮抗作用,从而易化阿片依赖和耐受的形成。因此阐明阿片抑制 视上核OT阳性神经元及OT释放的机制,对设计抗吗啡依赖和耐受形成的药 物有重要意义。...
