奥拉西坦对血管性痴呆大鼠认知功能的影响及对PI3K/Akt信号转导通路的作用

血管性认知功能障碍(vascular cognitive impairment,VCI)是由各种脑血管疾病引起的以学习、记忆功能障碍为主要症状的获得性智能损害综合征,其病程发展的晚期阶段被称作血管性痴呆(vascular dementia,VD)。目前,VD已经成为世界范围内继阿尔茨海默病(Alzheimer’s disease,AD)之后的第二大痴呆类型。但是,迄今为止,VD的发病机制尚不完全明确,因而缺乏有效的治疗药物。研究表明,慢性脑低灌注(chronic cerebral hypoperfusion,CCH)损伤在VD的发病过程中发挥着重要作用,因此探讨慢性脑低灌注损伤导致VD的发病机制,并寻求有效的痴呆防治药物成为现代医学的研究热点之一。已有的研究表明,神经元的自噬与凋亡在慢性脑低灌注损伤导致VD的发病机制中占有重要地位,因此调控神经元的自噬与凋亡对于减缓神经元损伤至关重要。磷脂酰肌醇-3-激酶/蛋白激酶B(phosphoinositide-3-kinase/protein kinase B,PI3K/Akt)信号通路是一条经典的抗凋亡、促存活通路,该通路的激活在缺血缺氧性神...
