CD147在双氢睾酮抗Aβ 42 诱发SH-SY5Y细胞损伤中的作用

阿尔茨海默病(Alzheimer’s disease,AD)是一种与年龄紧密相关的神经系统退行性疾病,临床上以记忆衰退和认知障碍为症状,病理上以脑内淀粉样蛋白(β-amyloid,Aβ)沉积、神经纤维缠结形成、神经元丢失等为特征。AD包括家族性和散发性两种,研究表明其病理机制可能与多种因素相关:Aβ沉积、离子通道、钙代谢紊乱、血管性异常、tau蛋白异常和氧化应激等。随着研究的深入发现Aβ在AD的发病进程中有着重要作用,认为Aβ的代谢异常打破了其生成与清除间的平衡关系,引发Aβ沉积。在Aβ生成与清除的动态过程中,多种酶类起到了关键作用,包括生成相关的β-、γ-分泌酶和降解相关的脑啡肽酶(Neprilysin,NEP)、胰岛素降解酶(Insulin degrading enzyme,IDE)、基质金属酶2/9(Matrix metalloproteinase2/9,MMP 2/9)等,共同维持着Aβ含量的平衡状态。有研究提示,睾酮下降可能是AD的一个危险因素,在AD之前或AD的早期就有可能出现。前瞻性研究中将AD的诊断与血睾酮水平的变化进行比较,对于首次临床诊断正常的男性调查对象随访4-3...
