【摘要】:目的:检测P-gp与HIF-1α在口腔鳞状细胞癌癌组织中的表达及相互关系,为临床评估口腔鳞癌化疗耐药性提供参考。方法:1、逆转录—多聚酶链反应(RT-PCR)检测口腔鳞癌癌组织P-gp(mdrl)基因及HIF-1 α基因在其中的表达。2、免疫组织化学SP法检测口腔鳞癌癌组织HIF-1α蛋白和P-gp蛋白在其中的表达。结果:1、28例OSCC癌标本,经测定符合实验要求的有15例。电泳结果:P-gp(mdrl)基因表达为15例,HIF-1α表达为13例。两者共表达有13例。列联相关性分析经卡方检验确切概率法得出结果,有统计学意义(P0.05)。两种基因在OSCC癌组织中的表达有正相关性,r=0.867。2、28例OSCC癌组织中HIF-1α蛋白阳性表达有21例(75%),P-gp蛋白阳性表达有20例(71.4%)。HIF-1α和P-gp表达在患者性别、肿瘤TNM分期、病理分级及淋巴结转移比较统计学无意义(P0.05)。HIF-1α蛋白表达与肿瘤大小(P=0.029)、年龄(P=0.041)存在差别,P-gp蛋白表达与肿瘤大小(P=0.007)存在差别,统计学有意义(P0.05)。28例OSCC组织中两者有阳性表达一致性共18例,列联相关性分析经卡方检验确切概率法得出结果,两者之间有统计学意义(P0.05),两者在OSCC癌组织中的表达有正相关性,r=0.548。结论:P-gp和HIF-1α在OSCC癌组织中同时表达增高,两者具有显著相关性,可能协同介导OSCC原发性耐药,其机制可能是通过HIF-1α上调P-gp表达来实现。P-gp和HIF-1α表达检测可为临床OSCC患者化疗耐药性评估提供参考指标。
[Abstract]:Objective: to detect the expression of P-gp and HIF-1 伪 in oral squamous cell carcinoma (OSCC) and their relationship, and to provide a reference for clinical evaluation of chemotherapeutic resistance in oral squamous cell carcinoma (OSCC). Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expression of P-gpmdrl) gene and HIF-1 伪 gene in oral squamous cell carcinoma (OSCC). Immunohistochemical SP method was used to detect the expression of HIF-1 伪 protein and P-gp protein in oral squamous cell carcinoma (OSCC). 15 cases met the requirements of the experiment and 15 cases were detected by electrophoretic analysis. The expression of HIF-1 伪 was found in 13 cases, and 13 cases were co-expressed. The results were obtained by the chi-square test and the exact probability method. There was a positive correlation between the expression of the two genes in OSCC tissues. There were 21 cases of positive expression of HIF-1 伪 protein in 28 cases of OSCC carcinoma. The positive expression of P-gp protein in 20 cases was 71.4%. HIF-1 伪 and P-gp were expressed in sex and TNM stage of tumor. There was no significant difference in pathological grade and lymph node metastasis between the expression of P0.05U. HIF-1 伪 protein and the size of the tumor (P0. 029, age: P0. 041). There was a significant difference between the expression of P-gp protein and tumor size (P0. 007). The positive expression of P-gp protein was consistent in 18 cases in 28 cases of OSCC. The results obtained by the exact probability method of chi-square test showed that there was a significant difference between the two methods and there was a positive correlation between them in OSCC cancer tissues. Conclusion the expression of HIF-1 伪 and P-gp in OSCC cancer tissues is increased simultaneously, which has a significant correlation, and may co-mediate the primary drug resistance of OSCC. The mechanism may be that the expression of. P-gp and HIF-1 伪 can be detected by up-regulating the expression of P-gp by HIF-1 伪, which may provide a reference index for the evaluation of chemoresistance in patients with OSCC.
