【摘要】:研究背景:口腔癌是一种常见的粘膜上皮性肿瘤,约占全身恶性肿瘤的3.5%,其发病率及死亡率不容忽视。其中发生于口腔粘膜的鳞状细胞癌约占口腔恶性肿瘤的90%。鳞状细胞癌是一种主要发生于黏膜、皮肤或附属器的上皮性恶性肿瘤,其恶性程度高,,转移率高,发病率高。因此,鳞状细胞癌成为医学领域亟待解决的难题。从基因水平研究肿瘤已成为研究者竞相追逐的焦点。近年来研究表明,全身恶性肿瘤的发生发展均与WAVE1、RAC1和CDK8基因的异常表达有关。因此,WAVE1、RAC1和CDK8基因越来越受到医学研究者的关注,成为目前研究肿瘤的新热点。 目的:观察在WAVE1、RAC1和CDK8口底鳞癌中的表达情况,探讨WAVE1、RAC1和CDK8在口底鳞癌治疗中和预后的意义。 方法:选择口腔鳞癌标本83例和43例正常口腔黏膜为对照组。采用免疫组化SuperPicTureTMPolyme法检测WAVE1,RAC1和CDK8的表达。并对口底癌组织WAVE1、RAC1和CDK8的表达进行相关性分析。 结果: 1. WAVE1、RAC1和CDK8在口底癌中的表达均高于癌旁组织的表达,差异有统计学意义。(P<0.05)。SP相关性分析显示,WAVE1,RAC1、CDK8的表达成正相关(P<0.05)。 2. WAVE1的表达与口底癌的组织分化及淋巴转移相关(P<0.05),与年龄及临床分期无关(p>0.05)。RAC1的表达与口底癌的组织分化、淋巴转移相关(P<0.05),与临床分期及年龄无关(p>0.05)。CDK8的表达与口底癌的组织分化及淋巴转移相关(P<0.05),与年龄及临床分期无关(p>0.05)。 结论: WAVE1,RAC1和CDK8在口底癌中呈现高表达。早期筛查WAVE1、 RAC1和CDK8指标对于口底鳞状细胞癌病情演变和预后评估有重要临床意义。
[Abstract]:Background: oral cancer is a common mucosal epithelial tumor. The incidence and mortality rate of malignant tumors of the whole body can not be ignored. The squamous cell carcinoma occurring in the oral mucosa accounts for about 90% of the malignant tumor of the mouth. Squamous cell carcinoma is a kind of mainly occurring in the mucous membrane. Epithelial malignant neoplasms of the skin or appendages with high malignancy, high metastatic rate, and high incidence. Squamous cell carcinoma (SCC) has become an urgent problem in the medical field. The study of cancer at the gene level has become a hot topic in recent years. The occurrence and development of malignant tumors of the whole body are related to the abnormal expression of RAC1 and CDK8 genes in WAVE1. therefore, the RAC1 and CDK8 genes of WAVE1 have been paid more and more attention by medical researchers and become a new focus of tumor research. Objective: to observe the expression of WAVE1pRAC1 and CDK8 in squamous cell carcinoma of the oral floor, and to explore the significance of WAVE1-RAC1 and CDK8 in the treatment and prognosis of squamous cell carcinoma of the floor of mouth. Methods: 83 cases of oral squamous cell carcinoma (OSCC) and 43 cases of normal oral mucosa were selected as control group. The expressions of WAVE1pRAC1 and CDK8 were detected by immunohistochemical SuperPicTureTMPolyme method, and the correlation between WAVE1pRAC1 and CDK8 was analyzed. Results:. 1. The expressions of RAC1 and CDK8 in the carcinoma of floor of mouth were higher than those in the adjacent tissues, and the correlation analysis showed that the expression of CDK8 was positively correlated with the expression of RAC1 and CDK8 in the tumor tissues (P < 0.05, P < 0.05, P < 0.05, P < 0.05). 2. The expression of WAVE1 was correlated with tissue differentiation and lymphatic metastasis of fundus oral carcinoma (P < 0.05), but not with age and clinical stage (P > 0.05). The expression of CDK8 was not correlated with the clinical stage and age, and the expression of CDK8 was not correlated with the histological differentiation and lymphatic metastasis of basal oral carcinoma (P < 0.05), but had no correlation with age and clinical stage (p > 0.05). Conclusion: high expression of WAVE1 / RAC1 and CDK8 is present in basal oral carcinoma, and early screening of WAVE1 / RAC1 and CDK8 is of great clinical significance in evaluating the prognosis and progression of squamous cell carcinoma of the floor of mouth.
