红景天苷对成骨细胞的作用及相关分子机制研究

【摘要】:目的:研究红景天苷(SAL)对人成骨样MG-63细胞增殖、分化和HIF-1α依赖的VEGF表达的影响及相关分子机制。方法:采用MTS法、流式细胞术分析、RT-PCR、ELISA、Western blot等技术观察SAL对MG-63细胞增殖、分化和HIF-1α依赖的VEGF表达的影响,进一步探讨其相关分子机制。结果:SAL通过调控MG-63细胞的周期分布及上调Runx2和Osterix表达促进成骨细胞的增殖和分化。另外,SAL可明显上调下游靶基因VEGF的表达。结论:SAL可显著促进成骨样MG-63细胞的增殖、分化以及HIF-1α依赖的VEGF的表达,进而促进骨和血管新生。
[Abstract]:Objective: to study the effect of salidroside on the proliferation, differentiation and HIF-1 伪 dependent VEGF expression of human osteoblast like MG-63 cells and its molecular mechanism. Methods: the proliferation of MG-63 cells was observed by MTS and flow cytometry. Differentiation and the effect of HIF-1 伪 -dependent VEGF expression, Results the cell cycle distribution of MG-63 cells was regulated and the expression of Runx2 and Osterix was upregulated to promote the proliferation and differentiation of osteoblasts. In addition, Sal could significantly up-regulate the expression of downstream target gene VEGF. Conclusion\% Sal can promote the proliferation and differentiation of osteoblasts by regulating the cell cycle distribution and upregulating the expression of Runx2 and Osterix. It can significantly promote the proliferation of osteoblast-like MG-63 cells. Differentiation and HIF-1 伪-dependent VEGF expression promote bone and angiogenesis.
