口腔鳞癌中CDK1的临床意义及相关miR-192的表达

【摘要】：目的:探讨细胞周期调控蛋白激酶1(cyclin-dependent kinases 1,CDK1)在口腔鳞癌患者中的临床意义及相关miR-192在口腔鳞癌中表达。方法:采用qPCR法检测8例口腔鳞癌组织中miR-192的表达,并分析其在口腔鳞癌中表达的趋势;同时用SP免疫组织化学测定77例口腔鳞癌标本CDK1蛋白的表达,并结合患者的术后复发、转移及5年生存率情况进行相关分析。实验结果:对比正常组织组,miR-192及pri-miR-192在口腔鳞癌中表达增加,miR-192更明显(P=0.0005160.01)。CDK1蛋白阳性表达与口腔鳞癌患者术后复发及淋巴转移关系密切,并有统计学意义(P0.05);同时,患者CDK1蛋白阳性组的5年生存率显著低于阴性组,两组之间有统计学意义(P0.05),阳性组患者预后更差。结论:评估口腔鳞癌患者CDK1蛋白的表达或许可作为预测口腔鳞癌患者预后的指标之一。miR-192在口腔鳞癌中表达增加,与正常组织相比有显著性差异;可进一步结合miR-192与CDK1共同预测口腔鳞癌的预后。
[Abstract]:Objective: to investigate the clinical significance of cyclin dependent kinases 1 (CDK1) in oral squamous cell carcinoma (OSCC) and the expression of miR-192 in oral squamous cell carcinoma (OSCC). Methods: qPCR method was used to detect the expression of miR-192 in 8 cases of oral squamous cell carcinoma (OSCC). At the same time, the expression of CDK1 protein in 77 cases of oral squamous cell carcinoma was detected by SP immunohistochemical method, and combined with the postoperative recurrence of the patients, the expression of CDK1 protein in oral squamous cell carcinoma (OSCC) was analyzed. Results: compared with the normal tissue group, the expression of miR-192 and pri-miR-192 in oral squamous cell carcinoma (OSCC) was significantly higher than that in normal tissue group (P < 0. 000516. 01). CDK1 protein expression was closely related to postoperative recurrence and lymphatic metastasis in patients with oral squamous cell carcinoma (OSCC). The 5-year survival rate in CDK1 positive group was significantly lower than that in negative group. Conclusion: the expression of CDK1 protein in patients with oral squamous cell carcinoma may be one of the predictors of prognosis of oral squamous cell carcinoma, and the expression of miR-192 is increased in oral squamous cell carcinoma. The prognosis of oral squamous cell carcinoma can be predicted by combining miR-192 and CDK1.
