AKT-Foxo3a信号通路调控小鼠MMP-20基因表达的研究

【摘要】:目的:本研究旨在探讨AKT-Foxo3a信号通路在基质金属蛋白酶-20(matrix metalloproteinase-20,MMP-20)基因表达中的作用。方法:分别通过免疫组化技术、实时定量RT-PCR技术以及双荧光素酶报告基因检测系统分析AKT-Foxo3a信号通路对MMP-20启动子转录活性的影响。结果:免疫组化染色结果显示,AKT及磷酸化Foxo3a(P-Foxo3a)均在分泌期成釉细胞核中呈阳性表达;RT-PCR检测结果显示,AKT阻断剂可显著抑制Foxo3a诱导的MMP-20基因的表达(P㩳0.05);双荧光素酶分析结果显示,AKT阻断剂亦可显著抑制Foxo3a调控MMP-20基因启动子的转录活性(P㩳0.05)。结论:转录因子Foxo3a可能通过AKT-Foxo3a信号通路调控MMP-20基因的表达,从而参与釉质发育过程。
[Abstract]:Objective: to investigate the role of AKT-Foxo3a signaling pathway in the expression of matrix metalloproteinase-20 (MMP-20) gene. Real-time quantitative RT-PCR technique and double luciferase reporter gene detection system were used to analyze the effect of AKT-Foxo3a signaling pathway on the transcriptional activity of MMP-20 promoter. Results: the results of immunohistochemical staining showed that the ameloblast in secretory phase was ameloblast with phosphorylated Foxo3a and phosphorylated Foxo3a. The results of reverse transcription-polymerase chain reaction (RT-PCR) showed that Foxo3a antagonist could significantly inhibit the expression of MMP-20 gene induced by Foxo3a. The results of double luciferase analysis showed that AK antagonist could significantly inhibit the transcriptional activity of MMP-20 promoter regulated by Foxo3a. Conclusion: the transcription factor Foxo3a may regulate the expression of MMP-20 gene through AKT-Foxo3a signaling pathway and thus participate in the development of enamel.
