牙龈卟啉单胞菌对血管内膜粘附因子和金属蛋白酶的影响

【摘要】:目的:通过给大鼠注射牙龈卟啉单胞菌(为牙周病的主要致病菌),观察其对血管内膜粘附因子和金属蛋白酶的影响,证实牙周疾病为动脉粥样硬化斑块形成的可能机制之一。 方法:120只大鼠随机分为对照组、低剂量组和高剂量组,各40只,肌注牙龈卟啉单胞菌(Pg),每3天注射一次,且均不进行抗生素干预,历时12周。取其静脉血,检测三组血液中的细胞间粘附分子-1、血管细胞粘附分子-1、基质金属蛋白酶-3及基质金属蛋白酶-9的水平。 结果:与对照组相比,低剂量组与高剂量组大鼠静脉血中ICAM-1[(175.79±14.30)ng/ml比(182.62±15.07)ng/ml.(189.39±14.93)ng/ml].VCAM-1[(256.49±37.17)ng/ml比(271.58±32.85)ng/ml.(286.66±30.66)ng/ml] MMP-3[3.23±0.69) ng/ml比(3.61±0.8)ng/ml.(3.97±0.83)ng/ml]及MMP-9[(1.30±0.39)mg/L比(1.48±0.39)mg/L、(1.67±0.45)mg/L]水平明显升高(P均0.05),且与低剂量组相比,高剂量组上述因子水平明显升高,差异有统计学意义(P0.05)。 结论:牙龈卟啉单胞菌会导致大鼠血清中细胞间粘附分子-1、血管细胞粘附分子-1、基质金属蛋白酶-3及基质金属蛋白酶-9含量的增加,而粘附分子可促进炎症的发生,基质金属蛋白酶可以降低弹性蛋白与胶原蛋白的含量,又可使包裹胆固醇的纤维帽分解、破裂,增加了冠心病的患病风险。进一步说明P.gingivalis的感染可以促进冠状动脉粥样硬化性心脏病。
[Abstract]:Objective: to investigate the effects of porphyromonas gingivalis (Porphyromonas gingivalis) on vascular intima adhesion factor (intima) and metalloproteinase (MMP) in rats, and to confirm that periodontal disease is one of the possible mechanisms of atherosclerotic plaque formation. Methods one hundred and twenty rats were randomly divided into control group, low-dose group and high-dose group, 40 rats in each group, injected intramuscularly with Porphyromonas gingivalis once every 3 days without antibiotic intervention for 12 weeks. The levels of intercellular adhesion molecule-1, vascular cell adhesion molecule-1, matrix metalloproteinase-3 and matrix metalloproteinase-9 were measured in three groups. Results: compared with the control group, ICAM-1 (175.79 卤14.30 ng / ml vs 182.62 卤15.07 ng / ml vs 182.62 卤15.07 卤14.93 ng / ml) .VCAM-1 [256.49 卤37.17 ng / ml vs 271.58 卤32.85 ng / ml vs 271.58 卤32.85 ng / ml] MMP-3 [3.23 卤0.69 ng/ml vs 3.61 卤0.83ngg. ml] and MMP-9 [1.30 卤0.39 mg / L = 1.48 卤0.39 mg / L] the level of MMP-3 [3.23 卤0.69 ng/ml vs 3.61 卤0.83ngg. ml] and MMP-9 [1.30 卤0.39 mg / L = 1.48 卤0.39 卤0.39 mg / L] significantly increased compared with the control group, and increased significantly compared with the above dosage group, and the dose was significantly higher than that in the control group (3.23 卤0.69 vs 182.62 卤15.07 ng / ml vs 182.62 卤15.07 卤14.93 卤14.93 ng / ml vs 271.58 卤32.85 ng / ml vs 271.58 卤32.85 ng / ml vs 3.23 卤0.69 vs 3.61 卤0.69 vs 3.61 卤0.83ngg. / ml) and MMP-9 (1.30 卤0.39 mg / L vs 1.48 卤0.39 mg / ml vs 271.58 卤32.85 ng / ml vs 271.58 卤32.85 ng / ml). The difference was statistically significant (P 0.05). Conclusion: porphyromonas gingivalis can increase the contents of intercellular adhesion molecule -1, vascular cell adhesion molecule -1, matrix metalloproteinase-3 and matrix metalloproteinase-9 in serum of rats. Matrix metalloproteinases reduce the content of elastin and collagen, and break down the fibrous cap that encapsulates cholesterol. The infection of P.gingivalis may promote coronary atherosclerosis heart disease.
