牙周病大鼠牙组织中MMP-9表达水平及与牙根吸收程度的关系

【摘要】:目的探讨牙周病大鼠牙组织中基质金属蛋白酶(MMP)-9表达与牙根吸收程度的关系。方法 12周龄健康雄性Wistar大鼠40只随机分为对照组和实验组,各20只。对照组仅给予正畸处理,实验组先建立牙周病模型,再给予正畸处理。分别于正畸牵拉前、牵拉1、2、4 w观察两组大鼠根尖吸收情况和牙组织MMP-9的表达情况。结果正畸牵拉前,两组大鼠均未见明显的根尖吸收情况;牵拉1 w后,两组大鼠均可见明显根尖吸收;在牵拉后2 w时,对照组根尖吸收活动基本停止,实验组继续增加,至4 w时基本停止。牵拉前,实验组的MMP-9阳性染色光密度值显著高于对照组(P0.05);牵拉后,两组大鼠的阳性染色光密度值均显著升高,对照组在牵拉后2 w开始下降,实验组在牵拉后4 w开始下降。两组大鼠的MMP-9阳性染色光密度值变化趋势与根尖吸收活动趋势基本一致,且实验组的光密度值显著高于对照组(P0.05)。结论牙周病大鼠牙组织中MMP-9的表达水平与牙根吸收活动的变化趋势基本一致,可作为判断牙根吸收的生物标志物之一。
[Abstract]:Objective to study the expression of matrix metal in rat periodontal disease periodontal tissue protease (MMP) the relationship between the -9 expression and the extent of root resorption. Methods 12 week old healthy male Wistar 40 rats were randomly divided into control group and experimental group, 20 rats in each. The control group only received orthodontic treatment, the experimental group to establish a model of periodontal disease then, given orthodontic treatment. In orthodontic traction, traction pull 1,2,4 w expression was observed in the two groups of rats MMP-9 and root resorption of periodontal tissue. The results of orthodontic traction, two groups of rats showed no obvious root resorption; stretch after 1 W, two groups of rats were root resorption; after pulling in 2 W, the control group root resorption activity basically stopped, the experimental group continued to increase to 4 W. Stop pulling, the experimental group MMP-9 positive staining optical density value is significantly higher than the control group (P0.05); after pulling, the two groups of rats were dyed the density values were significant With the increase in the control group after pulling 2 W began to decline, in the experimental group decreased after pulling 4 W. The two groups of rats MMP-9 staining optical density change trend and root resorption activity trend is basically the same, and optical density value of the experimental group was significantly higher than the control group (P0.05) and the expression level. The root tissue of rat tooth MMP-9 conclusion periodontal disease in the absorption change trend of activities are basically the same, can be used to determine the root resorption biomarkers.
