环孢素A诱导大鼠牙龈增生中CyPA,CD147,MMP-2,MMP-9的表达及意义

【摘要】:研究目的 本实验通过构建CIGO动物模型,通过免疫组织化学和Western blot的方法检测增生牙龈中CyPA、EMMPRIN(CD147)、MMP-2、MMP-9的表达,旨在探讨CIGO的发病机制。 研究方法 1.选取48只SD雄性大鼠,按随机数字表法分为CsA实验组和对照组,依据每日测量实际体重实验组给予CsA注射液腹部皮下注射,对照组给予适量蓖麻油皮下注射;从开始给药起,每周观察大鼠牙龈增生情况。 2.连续给药4周后,处死全部动物,运用影像学检查大鼠牙槽骨变化情况。 3.分离磨牙牙龈,运用组织学的方法观察牙龈的变化。 4.运用免疫组化的方法,检测CyPA、CD147、MMP-2、MMP-9在CsA实验组和对照组中的表达和定位。 5.运用Western blot的方法,检测CyPA、CD147、MMP-2、MMP-9四种蛋白在CsA实验组和对照组的表达量。 结果 1.大体观察结果:CsA实验组大鼠在连续注射CsA2周后,牙龈组织开始出现增生;经过CsA注射液的持续注射,4周后大鼠牙龈增生明显,牙龈外形如球状、结节状,隆起于牙齿表面,相邻牙龈乳头向牙冠方向增生明显,龈沟加深,,形成假性牙周袋,牙龈组织失去正常形态,牙周探诊未见出血,无牙周附着丧失;对照组牙龈未见增生。建模成功。 2. Micro CT检查结果:实验组与对照组大鼠均无牙槽骨丧失,牙槽骨高度无变化。 3. Western blot结果:CsA实验组中CyPA、MMP-2、MMP-9蛋白表达与对照组相比明显降低,但CD147蛋白表达在两组间无明显差异。 4.免疫组化结果:与对照组相比,CsA实验组中CyPA、MMP-2和MMP-9免疫组化阳性表达显著降低,差异具有统计学意义(P0.01);而CD147的阳性表达在两组间无明显变化,差异不具有统计学意义(P0.05)。 结论 CsA可能通过下调CyPA的表达,影响了CyPA与CD147的相互作用,使CyPA-CD147复合物介导的MMPs信号通路受到影响,下调了MMPs的表达,抑制了胶原的降解,引起胶原生成的相对增多,形成药物性牙龈增生。
[Abstract]:Research purpose. In this study, CIGO animal model was established, and the expression of MMP-9 in proliferative gingiva was detected by immunohistochemistry and Western blot. The aim of this study was to explore the pathogenesis of CIGO. Research method. 1. Forty-eight male SD rats were randomly divided into two groups: CsA group and control group. The experimental group was subcutaneously injected with CsA injection and the control group was subcutaneously injected with castor oil according to daily weight measurement. Gingival hyperplasia was observed weekly in rats. 2. After 4 weeks of continuous administration, all the animals were killed and the alveolar bone was examined by imaging. 3. The gingiva of molars was separated and the changes of gingiva were observed by histological method. 4. Immunohistochemical method was used to detect the expression and localization of MMP-9 in CsA group and control group. 5. Western blot was used to detect the expression of MMP-9 in CsA group and control group. Results. 1. The results of gross observation showed that gingival tissue began to proliferate after continuous injection of CsA2 for weeks in CSA group, and gingival hyperplasia appeared in rats after 4 weeks of continuous injection of CsA injection, the gingival shape was spherical and nodular, and it was protruding on the surface of teeth. The adjacent gingival papilla accreases to the crown direction obviously, the gingival groove deepens, forms the false periodontal bag, the gingival tissue loses the normal form, the periodontal diagnosis does not see the bleeding, has no periodontal attachment loss; the control group does not see the gingival hyperplasia. 2. Results of Micro CT: there was no alveolar bone loss and no change in alveolar bone height in the experimental group and the control group. 3. The Western blot results showed that the expression of CyPA-MMP-2MMP-9 in the CSA group was significantly lower than that in the control group, but there was no significant difference in the expression of CD147 protein between the two groups. 4. Immunohistochemical results: compared with the control group, the positive expression of CyPAA MMP-2 and MMP-9 in CSA group was significantly decreased, the difference was statistically significant (P 0.01), but the positive expression of CD147 had no significant change between the two groups, but the difference was not statistically significant (P 0.05). Conclusion. CsA may affect the interaction between CyPA and CD147 by down-regulating the expression of CyPA, affect the MMPs signaling pathway mediated by CyPA-CD147 complex, down-regulate the expression of MMPs, inhibit the degradation of collagen, and cause the relative increase of collagen production.
