肿瘤相关巨噬细胞和MMP-2在成釉细胞瘤中的表达及与血管生成的关系

【摘要】:目的:检测肿瘤相关巨噬细胞(tumor-associated macrophages,TAMs)和基质金属蛋白酶-2(matrix metalloproteinases-2,MMP-2)在成釉细胞瘤中的表达情况,探讨二者相关性及与成釉细胞瘤中微血管密度(microvessel density,MVD)的关系,初步探讨二者在成釉细胞瘤侵袭性生长中的作用机制。方法:收集成釉细胞瘤病理石蜡组织块43例作为研究对象,另收集正常口腔黏膜组织10例作为对照,采用免疫组织化学(Immunohistochemistry,IHC)通用二步法分别检测两组中CD68标记的巨噬细胞、MMP-2和CD34标记的微血管密度的表达情况,结果判定后统计学处理分析,进一步讨论。结果:1、TAMs在ABs中的计数为27.48±17.64,主要表达于肿瘤基质;MMP-2在所有ABs中均有不同程度表达,主要表达于肿瘤实质;MVD在ABs中的计数为21.84±8.20。2、TAMs、MMP-2和MVD在ABs中的表达均高于正常黏膜组织,差异有统计学意义(P0.01)。3、在ABs中,MMP-2表达和TAMs计数呈正相关关系(rs=0.350,P0.05),MVD计数随着TAMs和MMP-2的表达强度增加而增加,均成正相关关系(r=0.454,P0.01;rs=0.459,P0.01)。结论:1、TAMs和MMP-2在ABs中均有显著表达,参与ABs的发生发展;2、TAMs和MMP-2在ABs中的表达具有相关性,可能发挥协同作用;3、ABs中MVD较正常黏膜组织高,微血管生成参与肿瘤进展,TAMs和MMP-2可能发挥诱导血管生成作用促进ABs的侵袭性生长。
[Abstract]:Objective: to investigate the expression of tumor-associated macrophage tumor-associated macrophages (TAMs) and matrix metalloproteinases-2m-2 (MMP-2) in ameloblastoma, and to explore the relationship between them and microvessel density (MVD) in ameloblastoma.To explore the mechanism of the two in the invasive growth of ameloblastoma.Methods: 43 cases of ameloblastoma and 10 cases of normal oral mucosa were collected.The expression of MMP-2 and CD34 labeled microvessel density in macrophages labeled with CD68 in two groups were detected by Immunohistochemistry-IHC general two-step method. The results were analyzed by statistical analysis and discussed further.Results the count of Tams in ABs was 27.48 卤17.64, which was mainly expressed in the tumor matrix, MMP-2, in all ABs. The expression of MMP-2 and MVD in ABs were 21.84 卤8.20.2t in ABs. The expression of MMP-2 and MVD in ABs was higher than that in normal mucosa.There was a positive correlation between the expression of MMP-2 and the count of TAMs in ABs. The number of MVD increased with the increase of TAMs and MMP-2 expression, and the positive correlation was found between the expression of MMP-2 and the count of TAMs.Conclusion both Tams and MMP-2 were significantly expressed in ABs. The expression of Tams and MMP-2 in ABs was correlated with the occurrence and development of ABs, and the MVD expression in Tams was higher than that in normal mucosa.Microangiogenesis involved in tumor progression Tams and MMP-2 may play a role in angiogenesis and promote the invasive growth of ABs.
