NGF促进种植窝预备中下齿槽神经损伤后恢复的实验研究

【摘要】:目的 通过建立口腔种植体植入术中下齿槽神经损伤的动物实验模型,局部应用神经营养因子(NGF),促进种植手术过程中受损的下齿槽神经功能的恢复,从而为种植术中下齿槽神经损伤后的系统性治疗开辟一个新的思路。 方法 建立犬口腔种植术后下齿槽神经损伤动物实验模型,实验组于损伤后局部给予NGF微球,对照组给予空白微球,空白组不损伤神经,于1d、3d、7d、4w取神经损伤处远段制作标本,应用免疫荧光、Western Blot方法观察标本中NF-kappaB、GAP43的表达及神经生长情况。 结果 术后各组犬均成活,精神状态、饮食、活动正常,切口愈合良好,免疫荧光结果为空白组NF-kB阳性表达率低;对照侧术后1至3天后阳性表达率逐渐增加,7天时阳性表达率最高,至术后4周阳性表达率有所降低;实验侧术后1天后阳性表达率即增加,3天阳性表达率最高,并于7天至4周后阳性表达率逐渐下降。对照侧术后1至3天阳性表达率逐渐增加,7天阳性表达率最高,至4周后阳性表达率逐渐减弱;实验侧术后1到3天阳性表达率达到高峰,7天至4周阳性表达率迅速减弱。Western Blot结果显示NF-kB在空白组极少量表达;对照侧在术后1至3天显著表达,在7天达到峰值,,此后表达迅速下降,表达量均大于空白组;实验侧术后显著表达,3天达到峰值,此后有所下降,表达量均大于空白组。GAP43在空白组极少量表达;对照侧在术后1天和3天表达呈上升趋势,7天达到峰值,此后迅速下降,表达量均大于空白组;实验侧术后表达显著,3天达峰值,此后表达缓慢下降,表达量均大于空白组。 结论 神经生长因子缓释剂可为一种良好的修复周围神经损伤的蛋白,其临床前景广阔。
[Abstract]:Purpose By establishing the animal model of the injury of the inferior alveolar nerve in the dental implant implantation, the neurotrophic factor NGF was used locally to promote the recovery of the damaged inferior alveolar nerve function during the implant operation. Thus, a new idea for systematic treatment of inferior alveolar nerve injury in implant operation is opened up. Method The experimental model of the injury of the inferior alveolar nerve was established. The experimental group was given NGF microspheres locally after the injury, the control group was given the blank microsphere, the blank group did not injure the nerve, and the distal segment of the injured nerve was taken at 1 day, 3 days, 7 days and 4 weeks after the injury. The expression of NF-kappa Bon GAP43 and nerve growth were observed by Western Blot. Result All the dogs survived, mental state, diet, activity were normal, wound healing was good, the positive expression rate of NF-kB in the blank group was low, the positive expression rate of NF-kB in the control group increased gradually after 1 to 3 days after operation, the positive expression rate was the highest at 7 days after operation. The positive expression rate decreased at 4 weeks after operation, and the positive expression rate increased at 3 days after operation on the experimental side, and decreased gradually from 7 days to 4 weeks after operation. The positive expression rate of the control side increased gradually from 1 to 3 days after operation, the highest positive expression rate was on the 7th day, and the positive expression rate gradually decreased after 4 weeks. The positive expression rate of NF-kB decreased rapidly from 7 days to 4 weeks after operation on the experimental side. The results of Western Blot showed that the expression of NF-kB was very small in the blank group, while the positive expression rate in the control side reached the peak on the 1st to 3rd day after operation and reached the peak on the 7th day. After that, the expression of GAP43 decreased rapidly, and the expression of GAP43 was higher than that of the blank group, and the expression of GAP43 reached the peak 3 days after operation, and the expression of GAP43 was lower than that of the blank group. On the 1st and 3rd day after operation, the expression of the control side reached its peak value on the 1st and 3rd day after operation, and then decreased rapidly, and the expression level was higher than that in the blank group, and the expression reached the peak on the 3rd day after operation, and then the expression decreased slowly, and the expression was higher than that in the blank group. Conclusion Nerve growth factor (NGF) sustained-release agent is a good protein for repairing peripheral nerve injury.
