不同途径化疗诱导兔舌VX-2鳞癌转移淋巴结癌细胞凋亡研究

【摘要】:目的:研究经不同途径化疗对兔舌VX-2移植性鳞癌区域转移淋巴结癌细胞凋亡的影响。 方法:24只新西兰兔舌缘种植VX-2肿瘤,随机分为6组,每组4只。颈淋巴结最大横径大于5mm时开始化疗。A组:经舌动脉灌注紫杉醇脂质体和卡铂注射液;B组:经舌动脉灌注游离紫杉醇和卡铂注射液;C组:经舌动脉灌注5%葡萄糖溶液;D组:经耳缘静脉灌注紫杉醇脂质体和卡铂注射液;E组:经耳缘静脉灌注游离紫杉醇和卡铂注射液;F组:经耳缘静脉灌注5%葡萄糖溶液。治疗组给药剂量紫杉醇7.5mg/kg、卡铂13.0mg/kg。化疗后7天切除颈淋巴结进行检测:①透射电镜下观察转移灶凋亡细胞微结构的变化;②流式细胞仪检测淋巴结转移灶癌细胞凋亡率,ANOVA-LSD多重比较分析组间差异;③SP免疫组化检测P53蛋白表达情况,计数资料采用χ2检验和Fisher’s确切概率法。 结果:①各治疗组均可见明显的转移癌细胞凋亡。②动脉给药组癌细胞凋亡率高于静脉给药组;紫杉醇脂质体联合卡铂给药组凋亡率高于游离紫杉醇给药组。A组淋巴结缩小,,其余各组无缩小。各组癌细胞凋亡率分别为:A组(49.49±4.96)%、B组(29.65±3.61)%、C组(8.59±0.28%)、D组(21.05±1.65)%、E组(12.98±0.75)%、F组(8.47±0.30)%,除C组和F组外其余各组组间相比均有显著差异(P<0.05)。③P53蛋白在阳性率A组20.00%(2/12)、B组33.33%(4/12)、D组41.67%(5/12)、E组50%(6/12)、C组和F组75%(9/12),A组与C组和F组相比有显著差异(P<0.05),其他各组两两比较无统计学差异。 结论:靶动脉灌注较全身静脉灌注能更好诱导转移淋巴结癌细胞凋亡、抑制肿瘤细胞增殖。紫杉醇脂质体联合卡铂靶动脉灌注能有效诱导区域淋巴结转移癌细胞凋亡、抑制肿瘤细胞增殖。
[Abstract]:Aim: to study the effect of different ways of chemotherapy on apoptosis of metastatic lymph node cancer cells in transplanted squamous cell carcinoma (VX-2) of rabbit tongue. Methods Twenty four New Zealand rabbits with VX-2 tumor were randomly divided into 6 groups with 4 in each group. Group A: translingual infusion of paclitaxel liposome and carboplatin injection group B: translingual artery infusion of free paclitaxel and carboplatin injection group C: translingual artery infusion of 5% glucose Group D: paclitaxel liposome and carboplatin injection were perfused via auricular vein: group E: free paclitaxel and carboplatin injection were perfused via auricular vein: 5% glucose solution was perfused via auricular vein. The treatment group was given paclitaxel 7.5 mg / kg, carboplatin 13.0 mg / kg. Seven days after chemotherapy, cervical lymph nodes were resected to detect the changes of apoptotic microstructures of metastatic tumor cells under transmission electron microscope (TEM). 2 flow cytometry was used to detect the apoptosis rate of metastatic lymph node cancer cells and the difference of ANOVA-LSD multiplex analysis group was analyzed. The expression of p53 protein was detected by 3SP immunohistochemistry. The count data were analyzed by 蠂 2 test and Fisher's exact probability method. Results the apoptosis rate of metastatic cancer cells was significantly higher in arterial administration group than in intravenous administration group, and the apoptosis rate in paclitaxel liposome combined with carboplatin group was higher than that in free paclitaxel group.
