羧甲基壳聚糖温敏凝胶对NIH3T3细胞在病变牙根面附着和增殖的影响

【摘要】:目的:观察羧甲基壳聚糖(CMCS)温敏凝胶对病变牙根面NIH3T3细胞附着和增殖的影响。方法:60颗患牙制成60个牙根片,分为5组。A组:超声刮治+EDTA处理3 min+CMCS浸提液涂擦牙根表面5 min;B组:超声刮治+EDTA处理3 min+无菌生理盐水涂擦牙根表面5 min;C组:手动刮治+EDTA处理3 min+CMCS浸提液涂擦牙根表面5 min;D组:手动刮治+EDTA处理3 min+无菌生理盐水涂擦牙根表面5 min;E组(对照组):EDTA处理3 min+无菌生理盐水涂擦牙根表面5 min。将NIH3T3细胞与5组牙根片分别共培养16、24、72 h,采用细胞计数法和MTT法观察结果。结果:培养16 h时,A、B、C、D组细胞计数均高于对照组。培养24、72 h时,A、B、C、D组细胞增殖能力均高于对照组,且使用CMCS浸提液的A组及C组的细胞增殖能力高于使用无菌生理盐水的B组及D组(F=31.813和41.579,P0.05)。结论:采用手动或超声进行牙周刮治及根面平整均有助于NIH3T3细胞的附着;联合使用CMCS温敏凝胶处理牙根表面可以促进NIH3T3细胞在牙根面的附着与增殖。
[Abstract]:Aim: to observe the effect of carboxymethyl chitosan (CMCs) thermosensitive gel on the attachment and proliferation of NIH3T3 cells. Methods Sixty affected teeth were made into 60 root slices. Divided into 5 groups. Group A: ultrasonic scraping EDTA treatment 3 min CMCS extraction solution smeared root surface 5 minutes B group: ultrasonic scraping EDTA 3 min sterile saline smeared root surface 5 minutes C group: manual scraping EDTA treatment 3 min CMCS extraction Group D was treated with EDTA for 3 min and treated with normal saline for 3 min. Group E (control group) was treated with sterile saline for 3 min, and the root surface was smeared with normal saline for 5 minutes. The NIH3T3 cells were co-cultured with 5 groups of teeth for 24 ~ 72 h, and the results were observed by cell count and MTT method. Results: after 16 hours of culture, the cell counts in group D were higher than those in control group. The proliferative ability of group D was higher than that of control group, and the proliferative ability of group A and C with CMCS extract was higher than that of group B and D with aseptic normal saline and group D (F31.813 and 41.579P0.05N). Conclusion: manual or ultrasonic periodontal curettage and root surface leveling are beneficial to the attachment of NIH3T3 cells and the treatment of root surface with CMCS thermo-sensitive gel can promote the adhesion and proliferation of NIH3T3 cells on the root surface.
