人细胞分裂控制蛋白4在口腔鳞状细胞癌中的表达及临床意义

【摘要】:目的检测人细胞分裂控制蛋白4(hCDC4)在口腔鳞状细胞癌(OSCC)中的表达情况,分析其与临床病理参数之间的相关性。方法收集52例手术切除的OSCC组织及12例正常组织,运用免疫组织化学技术检测hCDC4表达情况,分析hCDC4蛋白表达与临床病理参数之间的相关性。应用hCDC4-siRNA瞬时转染OSCC细胞系Tca8113,MTT及蛋白免疫印迹(Western blot)检测细胞增殖能力及c-Myc和Cyclin E蛋白表达变化。结果 hCDC4蛋白在正常组织中的表达显著高于对应的OSCC组织(83.3%vs.25.0%,P0.05);hCDC4蛋白低表达与较大肿瘤直径(≥4cm)和高临床分期(Ⅲ+Ⅳ)显著相关(P0.05);敲低Tca8113细胞中hCDC4表达,导致细胞增殖能力显著增强(P0.05)以及c-Myc和Cyclin E蛋白蓄积。结论 hCDC4表达缺失导致c-Myc和Cyclin E蛋白蓄积可能是促进肿瘤进展的重要因素。
[Abstract]:Objective to detect the expression of human mitotic control protein 4hCDC4 in oral squamous cell carcinoma (OSCC) and to analyze its correlation with clinicopathological parameters. Methods Immunohistochemical technique was used to detect the expression of hCDC4 in 52 OSCC tissues and 12 normal tissues. The correlation between the expression of hCDC4 protein and clinicopathological parameters was analyzed. The cell proliferation and the expression of c-Myc and Cyclin E were detected by hCDC4-siRNA transient transfection into OSCC cell line Tca8113 and Western blot. Results the expression of hCDC4 protein in normal tissues was significantly higher than that in corresponding OSCC tissues (83.3vs.25.0).
