慢性咬合干扰疼痛模型中大鼠脑内孤啡肽表达的变化

【摘要】:目的建立大鼠单侧咬合紊乱的慢性咬合干扰模型,通过免疫组织化学方法,观察大鼠咬合干扰疼痛中脑N/OFQ(Nociceptin/Orphanin FQ)表达的变化,初步研究其在咬合干扰致咀嚼肌疼痛的中枢作用机制。方法本实验取8周龄SD大鼠24只,其中实验组建模共18只,在左侧上颌第一、二磨牙之间插入一正畸用橡皮筋(3 M U n i t e k,1/8#),使第一磨牙推向近中,逐渐形成上、下颌第一磨牙尖窝不吻合的咬合关系,实验期间每天检查橡皮筋有无脱落。对照组6只不施加咬合干扰,相同的状态下饲养。模型建立第5天,10天,21天后,随机抽取实验组各6只大鼠及第21天时对照组6只大鼠测量咀嚼肌痛阈值,然后麻醉灌注,取脑组织作切片,利用免疫组织化学方法检测大鼠中脑部分核团内N/OFQ的表达情况,初步探讨孤啡肽表达的改变与大鼠咬合干扰疼痛之间的关系。结果1.对照组的痛阈值为208.9±13,模型组第5天、10天、21天的痛阈值分别为117.8±11,73.3±10,198.7±6。2.免疫组化半定量分析结果显示,孤啡肽在对照组第21天时的平均光密度的表达为0.184±0.007;在模型组第5天、10天、21天的表达分别为0.253±0.008,0.326±0.010,0.203±0.004。任意两组比较差异均有统计学意义(P0.01)。孤啡肽在模型组中的表达均显著高于对照组,在模型组第10天的表达最高,在第21天的表达较第5天及第10天降低。结论咬合干扰模型中咀嚼肌的疼痛可促进大鼠脑内孤啡肽的表达;咀嚼肌痛觉敏感的程度与大鼠脑内孤啡肽的表达程正相关。
[Abstract]:Objective to establish a chronic occlusal interference model of unilateral occlusal disturbance in rats and to observe the changes in the expression of N/OFQ (Nociceptin/Orphanin FQ) in the pain of occlusal interference in rats by immunohistochemical method. The mechanism of the central action of the occlusal interference in the pain of masticatory muscles caused by occlusal interference was preliminarily studied. Methods 24 rats of 8 weeks old were taken from this experiment. The experiment was carried out in the experiment. A total of 18 models were set up, and a orthodontic rubber band (3 M U n i t e K, 1/8#) was inserted between the maxillary maxillary molars and the first molar was pushed to the near middle, and the occlusal relationship of the first molar of the mandibular first molar was gradually formed. The relationship between the pain of rat occlusion and pain was found. Results the pain threshold value of the 1. control group was 208.9 + 13, the model group was fifth days, 10 days, and the 21 day pain threshold was 117.8 + 11,73.3 + 6.2. immunohistochemical semi quantitative analysis results showed that the average light density of orphan enkephalin was 0.184 + 0.007 in the control group for twenty-first days, while in the model group, 10 days, 10 The expression of 21 days and 21 days was 0.253 + 0.008,0.326 + 0.010,0.203 + 0.004., and there were statistically significant differences between the two groups (P0.01). The expression of orphan enkephalin in the model group was significantly higher than that in the control group. The expression in the model group was the highest at tenth days, and the expression in the twenty-first day was lower than that in the fifth day and the 10 day. Pain can promote the expression of orphan enkephalin in rat brain, and the degree of masticatory myalgia sensitivity is positively correlated with the expression of orphan FQ in rat brain.
