咬合支持丧失对大鼠海马组织Bcl-2与Bax表达的影响

【摘要】:目的:观察咬合支持丧失大鼠脑组织海马区凋亡相关基因Bcl-2、Bax的表达,并分析其相关性。方法:选择四周龄SD大鼠50只,随机分为正常对照组、三颗磨牙缺失组、六颗磨牙缺失组、九颗磨牙缺失组与十二颗磨牙缺失组,每组各十只大鼠。咬合支持丧失组拔除相应牙数后常规饲养20周,然后取大鼠脑部海马组织。采用HE染色法观察各组脑组织海马区细胞形态结构改变,免疫组化染色法观察各组脑组织海马区Bcl-2、Bax阳性细胞表达。结果:正常对照组脑组织细胞形态结构正常,基本无损伤性改变。三颗磨牙缺失组细胞无明显变化;六颗磨牙缺失组出现细胞层数减少,结构排列紊乱;九颗磨牙缺失组海马组织神经细胞数目明显减少,细胞排列疏松;十二颗磨牙缺失组海马组织细胞内出现神经细胞数量锐减。咬合支持丧失组各组脑组织海马区Bcl-2阳性细胞数均较正常对照组显著减少,咬合支持丧失组各组脑组织海马区Bax阳性细胞数均较正常对照组显著增多(P均0.05)。结论:咬合支持丧失后脑组织海马区细胞启动了凋亡程序,并且与Bcl-2、Bax蛋白的表达存在相关性。
[Abstract]:Aim: to investigate the expression and correlation of apoptosis-related gene Bcl-2 (Bax) in hippocampus of rats with occlusal support loss. Methods: fifty four-week old SD rats were randomly divided into normal control group, three molar missing group, six molar missing group, nine molar missing group and twelve molar missing groups, each group had ten rats. The occlusal support loss group was fed for 20 weeks after extraction of the corresponding teeth, and then the hippocampus of the rat brain was removed. The morphological and structural changes of hippocampal cells in each group were observed by HE staining, and the expression of Bcl-2P Bax positive cells in hippocampus of each group was observed by immunohistochemical staining. Results: the morphology and structure of brain cells in normal control group were normal and no damage was found. There was no obvious change in the cells in the three molar loss groups, the cell layer decreased and the structure was disordered in the six molar missing groups, and the number of neurons in the hippocampal tissue decreased significantly in the nine molar absence groups, and the cell arrangement was loose. The number of nerve cells in the hippocampal tissue of the 12 molars missing group decreased sharply. The number of Bcl-2 positive cells in the hippocampus of the group with occlusal support loss was significantly lower than that in the control group, and the number of Bax positive cells in the hippocampus of the group with loss of occlusal support was significantly higher than that of the control group (P < 0.05). Conclusion: the apoptotic process was initiated in hippocampal cells after occlusal support loss and was correlated with the expression of Bcl-2 protein.
