年龄因素对牙周膜干细胞增殖和分化能力的影响

【摘要】:目的评估年龄因素对牙周膜干细胞(PDLSC)增殖、分化能力的影响。方法选取来源于12位不同的患者因阻生需要拔除的第三磨牙12颗,根据患者年龄分为A组(年龄18~20岁)、B组(年龄45~50岁)。分离培养获得PDLSC。MTT检测A、B组PDLSC增殖能力;进行成骨、成脂诱导,并用茜素红染色及油红O染色评价其分化能力;SA-βG染色分析A、B组细胞衰老情况;将A、B组PDLSC成骨诱导7 d后进行ALP染色并同时采用Western blotting检测OCN、ALP的表达。结果不同年龄阶段的供体中都能分离出PDLSC,然而其增殖和成骨分化能力随年龄增加而降低,成脂分化能力和SA-βG表达随年龄增加而增加。ALP和OCN成骨诱导7 d后表达水平均升高,且A组高于B组。结论随年龄增长能在牙周膜中获取PDLSC,随年龄增加其增殖与成骨分化能力下降,但成脂分化能力和SA-β表达则增加。
[Abstract]:Objective to evaluate the effect of age on the proliferation and differentiation of periodontal ligament stem cells (PDLSCs). Methods Twelve third molars from 12 patients were divided into group A (n = 18, 20 years old) and B group (n = 45 ~ 50 years old) according to the age of the patients. PDLSC.MTT assay was used to detect the proliferation of PDLSC, osteogenesis and lipogenesis were performed. The differentiation ability of PDLSC was evaluated by alizarin red staining and oil red O staining. ALP staining was performed 7 days after osteogenesis induction of PDLSC in Agna B group and the expression of OCN-ALP was detected by Western blotting. Results PDLSCs could be isolated from donors at different age stages. However, the proliferation and osteogenic differentiation ability of PDLSCs decreased with age, and the expression of SA- 尾 G and adipogenic differentiation increased with age. ALP and OCN increased after osteogenic induction for 7 days. Group A was higher than group B. Conclusion PDLSCs can be obtained from periodontal ligament with age, but the ability of adipogenic differentiation and SA- 尾 expression increase with age.
