微种植钉周期性加力压低犬磨牙牙周组织中MMP-2表达的实验研究

【摘要】:目的: 使用微种植钉压低犬磨牙,观察周期性加力不同时间牙周组织中MMP-2的表达,探讨微种植钉周期性加力压低犬磨牙,犬磨牙经长期压低后牙周组织的改建。 方法: 选取健康成年雄性纯种Beagle犬3只,于左侧上颌第二、三前磨牙颊、腭侧根分叉区各植入一枚微型种植钉,施加150g的力压低犬牙;每4周加力一次。左侧磨牙为实验组,随机分为三组,分别加力12、24、36周,右侧磨牙为空白对照。动物处死后,取上颌第二、三前磨牙及其周围牙周组织,固定、脱钙及制作组织学标本,切片后行HE染色及MMP-2免疫组化染色并光镜下观察,IPP软件计算MMP-2的平均光密度值,SPSS17.0软件包对数据进行统计学分析。 结果: HE染色光镜下观察显示,实验组较对照组组织学变化明显,,牙周膜宽度变窄,成纤维细胞增殖,随压低时间变化不明显;整个固有牙槽骨表面连续性破坏,加力12周,少量骨吸收陷窝主要出现在牙槽嵴区和根尖周区,加力24、36周,骨吸收陷窝较多,并见新骨沉积,成骨细胞排列在其表面;加力12周,根面牙骨质光滑,加力24周,根尖区牙骨质连续性破坏,出现牙骨质吸收陷窝,并有新生牙骨质覆盖,至加力36周,吸收陷窝接近根尖牙本质。 免疫组化染色结果显示,对照组牙周组织中MMP-2仅有少量表达;实验组,犬磨牙牙周组织中MMP-2的表达量明显升高,与对照组有显著性差异;压低24周牙周组织中MMP-2的表达量达最高;至压低36周表达量下降,但与对照组仍有显著性差异;压低12、24、36周组间相比差异均无显著性(P0.05)。 结论: 1.犬磨牙在长期压低过程中,牙槽骨改建发生在整个牙槽窝,以牙槽嵴顶部和根尖周区改建最为活跃。随着压低时间的延长,犬牙根尖吸收有逐渐加重的趋势。提示犬牙在长期压低过程中,根尖吸收与压低时间的长短有一定的相关性; 2.MMP-2主要表达在成纤维细胞、破骨细胞、成骨细胞以及骨细胞胞浆中,实验组表达量明显增加,进一步证实MMP-2参与了犬牙长期压低过程中牙周组织的改建; 3.牙周组织中MMP-2的表达量在实验组中明显增多,而没有随着压低时间的延长进一步升高。证实伴随微种植钉周期性地施加一定的压低力,经长期压低的犬磨牙牙周组织呈现一种动态平衡的骨改建过程。
[Abstract]:Objective:
The expression of MMP-2 in the periodontal tissues at different time was observed by the use of the micro implant nails, and the periodontal stress of the canine molar was lowered and the periodontal tissue of the canine molar was remodeled after a long period of depression.
Method:
3 healthy adult male purebred Beagle dogs were selected on the left maxillary second, third premolar buccal and the palatine lateral root bifurcate area. The force of 150g was applied to the lower canine teeth. The left molar was added every 4 weeks. The left molar was divided into three groups randomly, with a force of 12,24,36 weeks and the right molar as the blank control. After the animals were executed, taken from the death of the animals, the animals were executed, taken after the death of the animals, taken after the death of the animals, taken after the death of the animals. The maxillary second, third premolar and its surrounding periodontal tissue were fixed, decalcified and made tissue specimens. After the slice, HE staining, MMP-2 immunohistochemical staining and light microscopy were performed. The mean light density of MMP-2 was calculated by IPP software. The data were statistically analyzed by SPSS17.0 software package.
Result:
HE staining under light microscope showed that the histological changes of the experimental group were obvious, the width of the periodontal membrane was narrower, the proliferation of fibroblasts was not obvious with the low pressure time. The bone surface of the whole alveolar bone was continuously destroyed, and the bone resorption lacunae appeared in the alveolar ridge area and the periapical area for 12 weeks, and the bone resorption lacunae were added for 24,36 weeks, and the bone resorption lacunae were added. The osteoblasts were arranged on the surface of the osteoblast more, and the osteoblasts were arranged on the surface of the osteoblast for 12 weeks. The root surface of the cementum was smooth, and the cementum was continuously destroyed in the apical area for 24 weeks. The cementum resorption lacunae appeared, and the new cementum was covered to 36 weeks. The absorption of the lacunae was close to the root canine essence.
The results of immunohistochemical staining showed that only a small amount of MMP-2 was expressed in the periodontal tissue of the control group, and the expression of MMP-2 in the periodontal tissue of the experimental group was significantly higher than that in the control group. The expression of MMP-2 in the periodontal tissue was the highest in 24 weeks, and the 36 weeks' expression decreased, but there was a significant difference between the control group and the control group. There was no significant difference in 12,24,36 weeks between groups (P0.05).
Conclusion:
1. the alveolar bone remodeling occurred in the entire alveolar nest during the long period of depression, and the reconstruction of the alveolar ridge was most active in the top of the alveolar ridge and the periapical region. With the prolongation of the pressure, the root apex absorption of canine teeth increased gradually.
2.MMP-2 was mainly expressed in fibroblast, osteoclast, osteoblast and osteocyte cytoplasm, and the expression of the experimental group increased significantly, which further confirmed that MMP-2 was involved in the remodeling of periodontal tissue during the long-term depression of canine teeth.
The expression of MMP-2 in the 3. periodontal tissues was significantly increased in the experimental group, but not with the prolongation of the low pressure time. It was proved that the periodontal tissue of the canine molar with a long-term depression showed a dynamic and balanced bone remodeling process.
