基质胶缓释神经轴突导向分子SEMA3A对去势大鼠钛种植体稳定性的影响

【摘要】:目的:评估基质胶缓释SEMA3A对去势大鼠种植体稳定性的影响。方法:对60只SD大鼠行双侧卵巢切除术。12周后,根据植入种植体的不同将动物随机分为3组:1)对照组,种植体+蒸馏水;2)基质胶组,种植体+基质胶;3)种植体+基质胶+SEMA3A,将种植体植入所有动物胫骨近端的干骺端。在利用酶联免疫吸附试验检测种植体表面基质胶缓释SEMA3A的生物学过程进行检测。种植体植入后的第4周和第8周将胫骨与植入体一起收集,行显微CT、组织学和生物力学检测。结果:SEMA3A的体外释放检测发现,连续10d可以检测到基质胶中释放的SEMA3A,其中前3d呈爆发性释放过程;种植体植入后第4和第8周,与单独使用基质胶组相比,SEMA3A+基质胶组骨体积百分比分别增加了88.7%和83.3%,骨小梁粗度分别增加了51.4%和49.9%,骨小梁数量分别增加了54.2%和62.1%;在组织学定量评价中,种植体周围相对骨面积分别增加了89.8%和149.2%,骨与种植体结合率分别增加了148.9%和24.8%;生物力学检测发现,种植体的最大推出力增加了149.3%和209.2%,结合刚度增加171.2%和210.6%,结合韧性增加了151.8%和268.2%。结论:用SEMA3A加基质胶对种植体进行表面改性可以有效改善去势大鼠种植体的骨整合与稳定性。
[Abstract]:Objective: To evaluate the effect of matrix gel sustained release SEMA3A on the stability of implants in ovariectomized rats. Methods: after.12 weeks of bilateral oophorectomy in 60 SD rats, the animals were randomly divided into 3 groups according to the implantation implants: 1) the control group, the implant + distilled water; 2) matrix gel, implant + matrix glue; 3) implant + matrix glue +SEMA3A, which would implant the implant. The metaphysis of the proximal tibia of all animals was implanted. The biological process of detecting the sustained-release SEMA3A on the surface of the implant surface was detected by enzyme linked immunosorbent assay. The tibia was collected with the implant at fourth and eighth weeks after implantation. The microscopical CT, histology and Biodynamics test were performed. Results: the release of SEMA3A in vitro was detected. It was found that continuous 10d could detect the release of SEMA3A in matrix glue, in which the former 3D showed an explosive release process, and fourth and eighth weeks after implantation, the percentage of bone volume increased by 88.7% and 83.3% in the SEMA3A+ matrix group, respectively, and the bone trabecular size increased by 51.4% and 49.9% respectively, and the number of trabeculae increased respectively. In the quantitative evaluation of histology, the relative bone area around the implant was increased by 89.8% and 149.2%, and the bone and implant binding rate increased by 148.9% and 24.8%, respectively. The maximum release strength of the implant was increased by 149.3% and 209.2%, the stiffness increased by 171.2% and 210.6%, and the toughness was increased by 151.8% in combination with the toughness. And 268.2%. conclusion: the surface modification of the implant with SEMA3A plus matrix adhesive can effectively improve the osseointegration and stability of the implant in ovariectomized rats.
