老龄鼠实验性牙周炎的免疫特点

【摘要】:目的 :通过口腔涂抹牙周致病菌的方法建立小鼠牙周炎模型,比较低龄小鼠与老龄小鼠牙周炎发病免疫状态差异,分析Th17/Treg细胞相关因子在不同年龄牙周炎小鼠中的表达情况。方法:采用实体显微镜评价牙周骨吸收的程度,H-E染色法观察牙周组织病变的炎细胞浸润情况,TRAP染色法观察牙周骨组织表面破骨细胞浸润情况,实时定量PCR检测牙龈组织中Th17/Treg细胞相关因子TGF-β1、IL-10、IL-17A、RANKL m RNA的表达水平。采用SPSS17.0软件包对数据进行统计学分析。结果:实验4周后,牙周炎组与正常对照组相比,釉-牙骨质界至牙槽嵴顶的距离显著增加(P0.01),老龄组小鼠釉-牙骨质界至牙槽嵴顶的距离比低龄组小鼠显著增加(P0.01);牙周组织中大量炎细胞浸润,牙周袋加深;牙周组织中TGF-β1、IL-10 m RNA的表达水平显著降低(P0.01),IL-17A、RANKL m RNA表达水平显著增高(P0.01)。结论:老龄小鼠牙周炎中炎症介质的表达存在异常状态,比低龄小鼠牙周组织病损状态加剧,提示牙周炎的炎症状态可能不仅与其分泌的免疫抑制性细胞因子表达降低有关,更与其增龄性相关。
[Abstract]:Objective: to establish the periodontitis model of mice by oral smearing periodontal pathogenic bacteria, compare the difference of immune state of periodontitis between young mice and aged mice, and analyze the expression of Th17 / Treg cytokines in periodontitis mice of different ages. Methods: the degree of periodontal bone resorption was evaluated by solid microscope and the infiltration of inflammatory cells in periodontal tissue was observed by H-E staining and the infiltration of osteoclasts on the surface of periodontal bone tissue was observed by trap staining. The expression of TGF- 尾 1, IL-10, IL-17, RANKL mRNA in gingival tissues was detected by real-time quantitative PCR. SPSS 17.0 software package was used to analyze the data. Results: after 4 weeks of experiment, the periodontitis group was compared with the normal control group. The distance between enamel-cementum boundary and alveolar crest was significantly increased (P0.01), and the distance between enamel-cementum boundary and alveolar crest in aging group was significantly higher than that in younger group (P0.01), a large number of inflammatory cells infiltrated in periodontal tissue and periodontal bag deepened. The expression of IL-10 mRNA in periodontal tissues was significantly decreased (P0.01) and the expression of IL-17A1RANKL mRNA was significantly increased (P0.01). Conclusion: the expression of inflammatory mediators in periodontitis in aged mice is abnormal, which is more serious than that in young mice, suggesting that the inflammatory state of periodontitis may not only be related to the decreased expression of immunosuppressive cytokines secreted by periodontitis. It was also related to its aging.
