尼古丁对人牙龈成纤维细胞的增殖及IL-8表达的影响

【摘要】:目的:探讨尼古丁对体外培养人牙龈成纤维细胞(HGFs)的增殖及其分泌IL-8的影响。方法:组织块法原代培养HGFs,MTT法观察10、50、100、250、500、1000、2 000μg/mL不同浓度尼古丁对其增殖的影响;取第4代HGFs分别与10、50、100、250、500、1000μg/mL不同浓度尼古丁共同培养72 h,用酶联免疫吸附(ELISA)法检测HGFs培养上清中IL-8的含量。结果:50~2 000μg/mL不同浓度的尼古丁均能明显抑制HGFs的增殖(P0.05),且呈浓度依赖性;10~500μg/mL尼古丁均能明显促进HGFs合成IL-8(P0.05),其中以100μg/mL浓度组的促进作用最强。结论:一定浓度尼古丁可抑制HGFs的增殖并促进其合成IL-8,提示尼古丁可能通过调控IL-8的表达而造成对牙周组织的破坏。
[Abstract]:Aim: to investigate the effect of nicotine on (HGFs) proliferation and IL-8 secretion in cultured human gingival fibroblasts. Methods: the effects of different concentrations of nicotine at different concentrations on the proliferation of HGFs were observed by tissue block method, and the content of IL-8 in the supernatant of HGFs culture was detected by enzyme-linked immunosorbent (ELISA) (Elisa) for 72 hours after the fourth generation of HGFs was co-cultured with 1050,500,500,500 渭 g/mL nicotine for 72 hours. The results were as follows: (1) the content of nicotine in the supernatant of HGFs culture was determined by enzyme-linked immunosorbent (ELISA) (Elisa). Results nicotine at different concentrations of 渭 g/mL significantly inhibited the proliferation of HGFs (P0.05), and in a concentration-dependent manner, nicotine (10500 渭 g/mL) significantly promoted the synthesis of IL-8 by HGFs (P0.05), especially in the 100 渭 g/mL group. Conclusion: nicotine can inhibit the proliferation of HGFs and promote its synthesis of IL-8, suggesting that nicotine may damage periodontal tissue by regulating the expression of IL-8.
