有机硅季铵盐溶液浸泡后纯钛抗菌性能的研究

【摘要】:目的:本实验旨在研究经不同浓度的有机硅季铵盐溶液浸泡后纯钛磨片的抗菌性能以及抗菌持续时间。方法:1抗菌性实验完成打磨抛光、灭菌处理后,将120个钛片随机分为A、B、C、D、E、F六个大组,每组20片。6个大组又随机分为1、2、3、4四个亚组,每组5片。A组:采用1%浓度有机硅季铵盐溶液浸泡;B组:采用5%浓度有机硅季铵盐溶液浸泡;C组:采用10%浓度有机硅季铵盐溶液浸泡;D组:采用30%浓度有机硅季铵盐溶液浸泡;E组:采用60%浓度有机硅季铵盐溶液浸泡;F组:采用灭菌蒸馏水浸泡。四个亚组分别处理时间为:1组30 min,2组60 min,3组120 min,4组240min。处理完毕后取出放入灭菌蒸馏水中漂洗10min,取出,自然晾干后置入无菌24孔细胞培养板中。肺炎链球菌标准菌株ATCC49619复苏后,采用1640液体培养基进行稀释,浊度仪调整菌液浓度为0.5M。将不同分组的试件分别放入1ml菌悬液中浸泡,并放入37℃恒温箱中培养24h。取出试件,灭菌蒸馏水清洗试件上的浮菌,再将试件放入3ml1640液体培养基中,于振荡器上振荡1min,将液体培养基转移到比浊专用管中,混合均匀后使用浊度仪进行菌液浓度计数。采用无菌1640液体培养基作为空白对照。上述实验结果进行统计分析,发现1%有机硅季铵盐浸泡120min组为菌液浓度最低、抗菌性能最佳组,采取此种浸泡方式进行下面实验。2抗菌性持续时间实验选取纯钛试件30个,分为G、H、I、J、K五组,每组6个样本:G组经脱脂灭菌后采用1%有机硅季铵盐浸泡120min,将试件循环浸入5℃与55℃的恒温水浴中分别浸泡30s,循环0次后取出;H组经脱脂灭菌后采用1%有机硅季铵盐浸泡120min,将试件循环浸入5℃与55℃的恒温水浴中分别浸泡30s,循环2500次后取出;i组经脱脂灭菌后采用1%有机硅季铵盐浸泡120min,将试件循环浸入5℃与55℃的恒温水浴中分别浸泡30s,循环5000次后取出;j组组经脱脂灭菌后采用1%有机硅季铵盐浸泡120min,将试件循环浸入5℃与55℃的恒温水浴中分别浸泡30s,循环10000次后取出;k组仅采用脱脂灭菌处理,用蒸馏水进行浸泡处理。将处理好的各组试件分别放入1ml的0.5m菌悬液中,恒温箱37℃培养24h,取出试件,灭菌蒸馏水清洗试件上的浮菌,再将试件放入3ml1640液体培养基中,于振荡器上振荡1min,将液体培养基转移到比浊专用管中,混合均匀后使用浊度仪进行菌液浓度计数。仍采用无菌1640液体培养基作为空白对照。3数据处理采用spss13.0软件对本次研究所得数据进行统计学处理,选用kruskal-wallish非参数检验对抗菌性能进行分析,lsd检验进行两两比较,p0.05代表有统计学意义。选用dunnett-t检验对抗菌性时间实验数据进行统计学处理,p0.05代表有统计学意义。结果:1抗菌性测定结果不同浓度间的主效应存在差异,p0.05;但是对于不同处理时间之间的主效应无统计学意义,p0.05;两者之间存在交互作用,p0.05。对处理浓度因素进行分析比较结果显示:a、b、c、d、e、f这六组数据中,a组与b组,c组与d组、e组之间无差别;其余任意两组间数据比较均有差异p0.05。经过分析发现abcde五组与f组间数据有差异p0.05,说明a组1%浓度的处理能够让纯钛产生抗菌性。2抗菌性持续时间测定结果g组、h组、i组菌液浓度均低于k组,差异有统计学差异,p0.05;j组与k组之间进行比较无统计学差异,p0.05。结论:1有机硅季铵盐溶液浸泡纯钛后,能够让纯钛具备抗菌性能,其最适宜处理浓度为1%,最适宜处理时间为120min。2经1%有机硅季铵盐溶液浸泡纯钛120min后,其抗菌性能能够保持6个月左右。
[Abstract]:Objective: the purpose of this experiment was to study the antibacterial properties and duration of the pure titanium grinding tablets soaked with different concentrations of organosilicon quaternary ammonium salt solution. Methods: after 1 antibacterial experiments were finished, the 120 titanium slices were randomly divided into six groups, A, B, C, D, E, F, and each group of 20 pieces of.6 were randomly divided into four subgroups of 1,2,3,4, and were randomly divided into four subgroups of 1,2,3,4. Each group of 5 groups of.A groups were soaked in 1% concentration of silicone quaternary ammonium salt solution; group B: 5% concentration of silicone quaternary ammonium salt solution; group C: 10% concentration organosilicon quaternary ammonium salt solution; D group: 30% concentration organosilicon quaternary ammonium salt solution; E group: 60% concentration of silicon quaternary ammonium salt solution; F group: steeped distilled water soaked in sterilized water. The treatment time of the four subgroups was 30 min, 2 group 60 min, 3 group 120 min. 4 groups of 240min. were removed and rinsed into the sterile distilled water to rinse 10min, then put out, and then put into the sterile 24 hole cell culture plate after natural drying. The standard strain ATCC49619 of Streptococcus pneumoniae was resuscitation with 1640 liquid medium, and the turbidimeter adjusted the bacterial liquid. The concentration is 0.5M., the samples of different groups are soaked in 1ml bacteria suspension, and the specimens are incubated in the constant temperature box of 37 C, and the specimens are extracted by 24h., then the floating bacteria are cleaned by the sterilization distilled water. Then the specimens are put into the 3ml1640 liquid medium, and the oscillator oscillates 1min in the oscillator, and the Ye Tipei's foster group is transferred to the turbid special tube, and the mixture is used evenly after the use. The bacterial liquid concentration was counted by the turbidimeter. The aseptic 1640 liquid medium was used as the blank control. The results of the experiment were statistically analyzed. The results showed that 1% organosilicon quaternary ammonium salt soaked in 120min group was the lowest, and the best antibacterial performance group. This immersion method was used to test the antibacterial duration of the following experimental.2 for the selection of pure titanium specimen 30 G, H, I, J, K five groups, each group of 6 samples: G group after degreasing sterilization with 1% organosilicon quaternary ammonium salt soak 120min, immerse the specimen in 5 centigrade and 55 C in the constant temperature water bath respectively, respectively, after 0 times to take out, the H group after degreasing sterilization with 1% organosilicon quaternary ammonium salt soak 120min, immerse the test parts at 5 and 55 C constant temperature water The 30s was soaked in the bath for 2500 times, and the I group was soaked with 1% organosilicon quaternary ammonium salt for 120min after degreasing and sterilizing. The samples were soaked in 5 C and 55 C in the constant temperature water bath respectively for 30s, and then taken out for 5000 times. The j group was soaked with 1% organosilicon quaternary ammonium salt after degreasing and sterilizing, and the specimens were immersed in 5 and 55 degrees centigrade. 30s was soaked in the warm water bath and removed for 10000 times. The K group was treated with only degreasing sterilization and distilled water to soak. The treated samples were put into the 0.5m suspension of 1ml respectively. The incubators were incubated at 37 degrees centigrade for 24h, and the specimens were removed, and the floating bacteria were cleaned by sterilized distilled water, and then the specimens were put into the liquid medium of 3ml1640. The liquid medium was transferred to 1min, the liquid medium was transferred to the turbidimetric special tube, and the concentration of the bacteria was counted using the turbidimeter. The sterile 1640 liquid medium was still used as the blank control.3 data processing, and the data of this study were processed by the SPSS13.0 software, and the non parameter test of kruskal-wallish was selected. Analysis of antibacterial properties, LSD test for 22 comparison, P0.05 representative of statistical significance. Select the dunnett-t test for the antibacterial time experimental data processing, P0.05 representative of statistical significance. Results: 1 the main effect of different concentrations of antimicrobial determination, the main effect is different, P0.05; but for different treatment time There is no statistical significance between the main effect of P0.05 and the interaction between the two. The results of the analysis and comparison of the concentration factors of p0.05. show that there are no differences between the a group and the B group in the six groups of data of a, B, C, D, e and F, and there is no difference between the C group and the D group and the other two groups. The difference P0.05 shows that the treatment of a group 1% concentration can make pure titanium produce antibacterial.2 antibacterial duration determination result G group, H group and I group are lower than K group, the difference has statistical difference, P0.05, J group and K group have no statistical difference, p0.05. conclusion: 1 silicone quaternary ammonium salt solution after pure titanium, can make pure titanium. The optimum treatment concentration is 1%, the most suitable treatment time is 120min.2 after 1% organosilicon quaternary ammonium salt solution of pure titanium 120min, its antibacterial properties can be maintained for about 6 months.
