纤维连接蛋白修饰的微沟槽钛片对人牙龈成纤维细胞活性影响的研究

【摘要】:目的:研究微沟槽钛片表面经三氟代乙烷磺酰氯激活并修饰纤维粘结蛋白(fibronectin,FN)后对人牙龈成纤维细胞(HGF)活性的影响。方法:将FN修饰于经过激活的微沟槽钛表面,XPS分析材料表面元素组成;使用cck8法检测HGF接种材料表面6h、12h、1d、3d、5d、7d后的细胞增值率;荧光染色观察接种3d时的细胞数量及形态。I型胶原纤维ELISA实验及Real-time PCR检测接种5d时材料表面对HGF分泌功能的差异。结果:在实验观察的6个时间段实验组材料细胞增值率高于对照组(P0.05)。3d时HGF在实验组材料表面的伸展率高于空白组(P0.05)。5d时FN修饰的两组材料上的细胞外I性胶原纤维的数量及mRNA的表达水平较空白组高(P0.05)。结论:经三氟代乙烷磺酰氯处理修饰FN的微沟槽钛片可促进HGF的增殖、伸展及分泌功能。
[Abstract]:Aim: to study the effect of activated and modified fibronectin FN on the (HGF) activity of human gingival fibroblasts on the surface of microgrooved titanium tablets by trifluoroethanesulfonyl chloride. Methods: FN was modified on the activated microgrooved titanium surface to analyze the surface element composition, and the cell proliferation rate of the HGF inoculated material was detected by cck8 method after 6 hours, 12 hours, 1 day, 3 days and 5 days after inoculation. The number and morphology of type I collagen fibers at 3 days after inoculation were observed by fluorescence staining, and the difference of HGF secretion on the surface of the material was detected by Real-time PCR after 5 days inoculation. Results: the proliferation rate of HGF on the material surface of the experimental group was higher than that of the control group (P0.05). 3 days after the experiment, the extension rate of HGF on the material surface of the experimental group was higher than that of the control group (P0.05). 5 days after FN modification, the extracellular collagen fibers were higher in the experimental group than in the control group (P0.05). The quantity of mRNA and the expression level of mRNA were higher than those of the blank group (P0.05). Conclusion: the microgrooved titanium tablets modified with trifluoroethanesulfonyl chloride can promote the proliferation, extension and secretion of HGF.
