MALDI-TOF质谱技术鉴定龋病患者唾液中的变异链球菌

【摘要】:目的:利用基质辅助激光解吸电离时间飞行质谱(matrix-assisted laser desorption/ionization time-of-flight mass spectrometry,MALDI-TOF MS)鉴定龋病患者唾液中的变异链球菌,为探寻快速、准确鉴定变异链球菌的方法提供依据。方法:研究对象为北京大学口腔医院牙体牙髓科就诊的90名龋病患者,收集受试者的唾液样本,处理后进行MALDI-TOF MS检测,结果导入BioExplorer 1.0软件与标准库比对,分值≥25者鉴定为变异链球菌,25者鉴定为非变异链球菌。最后将质谱和16S rDNA测序结果进行比对,计算灵敏度和一致率。结果:MALDI-TOF MS鉴定变异链球菌的灵敏度为96.0%,与16S rDNA测序结果的一致率为98.7%。结论:MALDI-TOF MS是一种高通量、快速且操作简便的方法,鉴定变异链球菌的灵敏度和一致率都很高,具有较高的临床应用价值。
