犬骨髓间充质干细胞复合磷酸钙骨水泥修复骨缺损的实验研究

【摘要】:目的 探讨犬骨髓间充质干细胞复合磷酸钙骨水泥支架材料修复骨缺损效果。为口腔种植修复中骨组织缺损的修复与重建提供新思路与方法。 方法 分离培养犬骨髓间充质干细胞(BMSCs),观察其成骨诱导作用;将细胞与磷酸钙骨水泥支架材料(CPC)复合用于动物实验;在4只比格犬的下颌骨每侧制造3处大小相同的骨缺损。随机将骨缺损分为三组:BMSCs-CPC组(移植复合种子细胞的支架材料),CPC组(只移植支架材料),空白组(不做任何处理)。移植后分别于4周、8周各处死两只犬,大体、X线、组织学观察不同时期各组骨缺损的成骨情况。 结果 骨髓间充质干细胞成骨诱导14天后茜素红染色可见钙化结节形成,,碱性磷酸酶染色细胞胞浆出现深色颗粒,表明骨髓间充质干细胞向成骨细胞分化;术后4、8周各组骨缺损均有不同程度的骨再生,BMSCs-CPC组、CPC组中骨组织再生情况优于空白组。BMSCs-CPC组中支架材料降价情况,新骨形成数量,骨组织的成熟程度优于CPC组。 结论 骨髓间充质干细胞是一种具有多向分化潜能和高度增殖能力的干细胞,可以向成骨细胞分化。磷酸钙骨水泥支架材料具有良好的生物相容性,对于维持骨外形加快成骨过程起到良好的作用。利用组织工程的原理,BMSCs种子细胞复合CPC支架材是一种有效的、促进新骨再生的骨缺损修复方法,有利于颌骨高度及宽度的保存,为后期口腔种植修复提供了有益条件。
[Abstract]:Objective to investigate the effect of bone marrow mesenchymal stem cells (MSCs) combined with calcium phosphate cement scaffold for repairing bone defect. To provide a new idea and method for the repair and reconstruction of bone tissue defect in dental implant repair. Methods (BMSCs), was isolated from cultured canine bone marrow mesenchymal stem cells (BMSCs) to observe its osteogenic induction, and the cells were combined with calcium phosphate cement scaffold (CPC) for animal experiment. Three bone defects of the same size were made on each side of the mandible of 4 Beagle dogs. Bone defects were randomly divided into three groups: BMSCs-CPC group (), CPC group) and blank group (no treatment). Two dogs were killed at 4 and 8 weeks after transplantation. The osteogenesis of bone defects in each group was observed by gross X-ray and histology. Results 14 days after osteogenesis of bone marrow mesenchymal stem cells, calcified nodules were found in alizarin red staining, dark granules appeared in the cytoplasm of cells stained with alkaline phosphatase, which indicated that bone marrow mesenchymal stem cells differentiated into osteoblasts. Bone tissue regeneration in BMSCs-CPC group and CPC group was better than that in blank group, but in BMSCs-CPC group, the reduction of scaffold material, the number of new bone formation and the maturity of bone tissue were better than those in CPC group. Conclusion Bone marrow mesenchymal stem cells are a kind of stem cells with multiple differentiation potential and high proliferative ability, which can differentiate into osteoblasts. Calcium phosphate cement scaffold has good biocompatibility and plays a good role in maintaining bone shape to accelerate osteogenesis. Based on the principle of tissue engineering, BMSCs seed cells combined with CPC scaffold is an effective method for repairing bone defect, which is beneficial to the preservation of the height and width of jaw bone, and provides a beneficial condition for the later stage of oral implant repair.
