根尖周损伤后根管内组织的变化过程及基质细胞衍生因子1释放的实验研究
[Abstract]:Objective: to observe the changes of tissue and stromal cell derived factor 1 (stromal cell derived factor-1,) in the root canal of maxillary first molar induced by SD by stimulating periapical hemorrhage. SDF-1) expression in root canal and the change of serum SDF-1 concentration before and after injury. To investigate the role of SDF-1 in dental pulp vascular regeneration. Methods: 64 SPF grade SD male rats of 6-8 weeks old, weight 180-220 g. The left maxillary first molar model of unmyelinated root canal was established and the periapical injury was carried out in the experimental animals (56 rats). The injury group was randomly divided into 7 groups according to different time points after periapical injury. The animals were killed and blood and maxillary bone were collected after anesthesia. Enzyme linked immunosorbent assay (enzyme linked immunosorbent assay,ELISA) was used to detect the content of SDF-1 in serum. The changes of root canal tissue and the expression of SDF-1 in root canal were observed by histochemical staining after maxillary fixation and decalcification. The results showed that the blood clotting in the root tip of the root canal showed fibrin in the blood clots and blood cells in the root canal at 2 and 4 days after the injury. In the late stage of tissue injury, hemagglutination and inflammatory cells in the root canal decreased, and the connection between tissue and dentin was close. On the 14th day after injury, neovascularization appeared in the tissue and cell structure appeared between the tissue and the root canal wall. Immunohistochemical staining showed that the expression of SDF-1, in blood clots and inflammatory cells in root canal after trauma increased with time, blood clots and inflammatory cells decreased, and SDF-1 expression decreased. ELISA detection showed that after periapical injury, the expression of SDF-1, was decreased. The serum SDF-1 concentration in 12h group and 4-day group was significantly higher than that before injury (P0.05). Conclusion: 1. The process of pulp vascular regeneration in root canal and the expression of SDF-1 in root canal were related to blood clots and inflammatory cells after apical hemorrhage. 2. The concentration of SDF-1 in serum increased at 12 h and 4 d after periapical injury, but it could not be sustained in the whole process of pulp vascular regeneration. The evidence of blood circulation homing by stimulating periapical injury to chemotaxis of bone marrow stem cells could not be confirmed by further experiments.
