干扰素刺激基因在人牙髓组织及牙髓细胞中表达的研究

【摘要】:目的:研究干扰素刺激基因(STING)在人牙髓组织和细胞中的表达。方法:分别采用逆转录-聚合酶链反应(RT-PCR)检测STING在体外培养的人牙髓细胞中的表达,免疫细胞化学和免疫组化技术检测STING在人牙髓组织的表达和定位。结果:STING mRNA在人牙髓细胞中有显著表达;而在牙髓组织中,STING主要表达于牙髓组织表面的成牙本质细胞的胞浆中。结论:在人牙髓组织中,STING在基因和蛋白水平均呈高表达,且主要定位于成牙本质细胞的胞浆中;提示STING调节通路可能对宿主的天然免疫反应发挥调节作用。
[Abstract]:Objective: to study the expression of interferon stimulating gene (STING) in human dental pulp tissues and cells. Methods: the expression of STING in human dental pulp cells was detected by reverse transcription-polymerase chain reaction (RT-PCR), and the expression and localization of STING in human dental pulp tissue were detected by immunocytochemistry and immunohistochemistry, respectively. Results: STING mRNA was significantly expressed in human dental pulp cells, while STING was mainly expressed in the cytoplasm of odontoblast cells on the surface of dental pulp tissue. Conclusion: in human dental pulp tissue, STING is highly expressed at gene and protein levels, and is mainly located in the cytoplasm of odontoblast cells, suggesting that STING regulatory pathway may play a regulatory role in the innate immune response of the host.
