【摘要】：目的:本研究是根据我省历年来上报的乙型脑炎资料分析我省乙型脑炎的流行趋势,并在不同生态环境中捕捉蚊虫标本,找出我省不同生态环境中的优势蚊种,同时用细胞培养的方法从蚊体内分离病毒,并用常规PCR和荧光PCR的方法进行初步鉴定,筛选乙型脑炎病毒,为以后对病毒进行核苷酸序列分析、鉴定病毒种类、研究我省存在的乙脑病毒与国内、外其他地域同种病毒的差异,绘制系统发生树,进而研究病毒的进化规律和分析目前使用的疫苗有无保护作用打下基础。方法: 1.资料收集:根据历年来我省上报的乙型脑炎资料进行统计,参考我省已发表的有关此课题的文献,进行分析乙脑流行趋势及其原因。 2.标本的收集运输:在不同生态环境中将捕到蚊子,快速冻死后剔除雄蚊,将蚊子分类,50～100只为一组,装入冻存管中,用医用胶布封口,并用记号笔编号,冻存管放入布袋中后及时放入液氮中,送至实验室。 3.根据在我省不同生态环境中采集蚊虫标本结果,分析我省不同生态环境
[Abstract]:Objective: to analyze the epidemic trend of Japanese encephalitis in our province according to the data of Japanese encephalitis reported over the years, and to capture mosquito specimens in different ecological environment and find the dominant species in different ecological environment in our province, and divide the virus from the mosquito in the body with the method of cell culture, and use the conventional PCR and fluorescence PCR methods. Preliminary identification, screening of Japanese encephalitis virus, in order to analyze the nucleotide sequence of the virus and identify the species of the virus in the future, and study the difference between the virus of the encephalitis B and the domestic and other regions of the same virus, draw the phylogenetic tree, and then study the evolution of the virus and analyze the protection of the current vaccine.
Method:
1. data collection: according to the statistics of Japanese encephalitis data reported in our province over the years, and referring to the published literature on this subject in our province, the trend of epidemic encephalitis B and its reasons are analyzed.
The collection and transportation of 2. specimens: the mosquitoes will be caught in different ecological environment, the mosquitoes are removed after quick freezing and the mosquitoes are removed. The mosquitoes are classified, 50~100 are one group, they are loaded into the cryopreservation tube, sealed with the medical glue cloth, and numbered with the marker pen. The cryopreservation pipe is put into the liquid nitrogen and sent to the laboratory in time after the storage tube is put into the cloth bag.
3. according to the results of collecting mosquito specimens in different ecological environment of our province, we analyze the different ecological environment in our province.
