具有抑制钉螺体内酶活性的天然化学成分的研究

【摘要】: 血吸虫病是我国十一五期间提出的四大重点控制疾病(艾滋病、结核病和乙肝)之一,是我国危害最为严重的寄生虫病,自从2005年首次报道空心莲子草浸提液含有抑制钉螺运动的化学成分后,我们(武汉科技大学和贵州大学)发现用全草干粉的浸提液(1g/100ml水)可在短时间内使钉螺失去运动能力,本课题继续在前一阶段的研究成果的基础上,对空心莲子草中所含有的有效化学成分进行提取分离、纯化。 空心莲子草Alternanthera philoxeroides(Mart.)Griseb.,为苋科莲子草属植物,具有清热、凉血、解毒等功效,用于治疗麻疹、流行性感冒等病毒性疾患。本文的研究内容和结果如下: 研究空心莲子草抑制钉螺运动的机理:将实验钉螺随机分为4组,分别于空心莲子草浸液(1 g/L)及去氯水中各浸泡12 h和20 h后,用酶组织化学方法染色,显微镜下观察钉螺的头足部、中枢神经节、鳃叶及肝脏的三磷酸腺苷酶(Mg~(2+)-ATPase)、胆碱脂酶(ChE)、乳酸脱氢酶(LDH)和琥珀酸脱氢酶(SDH)的活性;并用计算机图像分析系统(HPIAS-1000)检测各组不同部位染色片的灰度值,进行定量分析。得出结论空心莲子草浸液主要通过迅速抑制钉螺体内ChE活性,随后抑制Mg~(2+)-ATPase活性,而导致ATP的释放与利用障碍致钉螺死亡。 对苋科莲子草属植物空心莲子草的化学成分进行研究:空心莲子草干燥后粉碲,经0.1mol/L HCl渗漉提取,石油醚萃取,用氨水调酸水层pH≈12,经氯仿萃取,得氯仿层浸膏,浸膏反复经硅胶柱色谱、葡聚糖凝胶(Sephadex LH-20)柱色谱、反相硅胶(RP-18)柱色谱分离纯化得单一成分,结构用现代波谱分析技术(MS、NMR、HPLC等)和理化方法鉴定。从该种植物中分离得到一个单一成分为生物碱,鉴定为β-carboline(APG-2)。得出结论为化合物APG-2为首次从该植物中分离得到。
[Abstract]:Schistosomiasis is one of the four major diseases (AIDS, tuberculosis and hepatitis B) put forward during the 11th Five-Year Plan in China. It is the most serious parasitic disease in China. Since 2005, when it was first reported that the extract of lotus seed contains chemical constituents that inhibit the movement of Oncomelania hupensis, We (Wuhan University of Science and Technology and Guizhou University) found that the extraction solution (1g/100ml water) of whole straw dry powder can make snail lose movement ability in a short time. The active chemical constituents of lotus seed were extracted and purified. Alternanthera philoxeroides (Mart.) Griseb., is a plant of the genus Lotus Seeds in Amaranaceae. It has the functions of clearing heat, cooling blood and detoxifying. It is used to treat viral diseases such as measles, influenza and so on. The research contents and results are as follows: to study the mechanism of inhibition of Oncomelania hupensis movement by lotus seed grass: the experimental snails were randomly divided into 4 groups. The snails were soaked in lotus seed extract (1 g / L) and dechlorinated water for 12 h and 20 h, respectively. The head, foot and central nervous ganglion of Oncomelania hupensis were observed under microscope by enzyme histochemical staining. Activities of Mg~ (2)-ATPase), cholinesterase (ChE), lactate dehydrogenase (LDH) and succinate dehydrogenase (SDH) in Gill leaves and liver; Computer image analysis system (HPIAS-1000) was used to detect the grayscale value of each group. Conclusion the results showed that the extract of lotus seeds could rapidly inhibit the activity of ChE in Oncomelania hupensis, and then inhibit the activity of Mg~ (2)-ATPase, which resulted in the release and utilization of ATP and resulted in the death of Oncomelania hupensis. In this paper, the chemical constituents of Hollow Lotus Seeds of Amaranaceae were studied.
