【摘要】:目的:观察术后肠梗阻对大鼠脑内nesfatin-1表达的影响。 方法:大鼠经腹腔注射硫酸仲丁巴比妥(100~150mg/kg)麻醉后行腹部手术,术后2小时后经心灌注固定,免疫组化观察大鼠脑内c-Fos和nesfatin-1免疫阳性神经元的表达。 结果:手术组大鼠下丘脑视上核(SON)(t=6.9,p0.01),室旁核(mpPVN(t=17.2; p0.01), mmPVN (t=25.8, p0.01),1mPVN (t=19.5; p0.01)和apPVN(t==22.1;p0.01)),蓝斑(LC)(t=2.6,p0.05),动眼神经副核(EW)(t=10.74, p0.01),中缝苍白核(rRPa)(t=2.57,p0.05),孤束核(NTS)(t=10.46,p0.01),延髓腹外侧区(VLM)(t=7.86,p0.01)神经元c-Fos表达显著增加,但nesfatir-1免疫阳性神经元表达无显著改变(P0.05)。但统计分析发现,腹部手术激活的c-Fos神经元中,在下丘脑SON内99%、apPVN内71%、1mPVN内47%、mmPVN内41%以及mpPVN内9%表达nesfatin-1;在脑干,腹部手术可分别激活LC内91%、rRPa内82%、EW和VLM内74%、NTS14%表达c-Fos的iesfatin-1神经元。 结论:腹部手术可激活下丘脑和脑干摄食相关中枢内nesfatin-1神经元,其对摄食和胃排空障碍疾病调控具有潜在临床意义。
[Abstract]:Objective: To observe the effect of intestinal obstruction after operation on the expression of nesfatin-1 in the brain of rats.
Methods: abdominal operation was performed after intraperitoneal injection of SEC butyl barbiturate (100 ~ 150mg/kg). After 2 hours, the expression of c-Fos and nesfatin-1 immunoreactive neurons in rat brain was observed by immunohistochemistry.
Results: the hypothalamic supraoptic nucleus of rats in operation group (SON) (t=6.9, P0.01), paraventricular nucleus (mpPVN (t=17.2; P0.01), mmPVN (t=25.8, P0.01), 1mPVN (t=19.5; P0.01) and apPVN (t==22.1; P0.01)), blue (LC) (t=2.6, P0.05), oculomotor nerve accessory nucleus (EW) (t=10.74, P0.01), nucleus raphe pallidus (rRPa) (t=2.57, P0.05), the nucleus of the solitary tract (NTS) (t=10.46, P0.01), ventrolateral medulla (VLM) (t=7.86, P0.01) expression of c-Fos neurons increased significantly, but the expression of nesfatir-1 immunoreactive neurons did not change significantly (P0.05). But the statistical analysis found that the activation of c-Fos neurons in abdominal surgery, SON in the hypothalamus in 99%, apPVN in 71%, 1mPVN in 47%, mmPVN 41% and mpPVN 9% expression of nesfatin-1; in the brainstem, abdominal surgery can activate LC in 91%, rRPa in 82%, EW and VLM in 74%, NTS14% expression of c-Fos iesfatin-1 neurons.
Conclusion: abdominal operation can activate nesfatin-1 neurons in hypothalamic and brainstem feeding related central nervous system, and it has potential clinical significance in regulating the disease of ingestion and gastric emptying disorders.
