氢氧化钠介入法建立病态窦房结综合征大鼠模型及细辛对其心率的影响

【摘要】:目的:(1)为研制出一种简单精确损伤小的病态窦房结综合征大鼠模型,进一步改进造模方法,简化实验操作,提高造模成功率;(2)研究细辛对病态窦房结综合征模型心率的影响;(3)探讨细辛作用于病态窦房结综合症大鼠模型的机制,分析其在病窦综合征中对心率影响的机理。 方法:(1)使用心电筛查健康大鼠,择40只HR在350-450次/分的大鼠。乙醚麻醉后,于大鼠胸骨右缘第二肋间处皮肤剪一约1毫米小口,并于开口周围作预荷包缝合穿线。将自制标测电极导管经由该口紧贴胸骨右缘第2肋间插入,给药部位根据心电图波形来确定,使用微量注射器经自制标测电极导管缓缓推注氢氧化钠,进行局部浸润以建立sSS大鼠模型,最后进行完整的荷包缝合。(2)从40只大鼠病态窦房结综合征模型中,选取32只成功模型大鼠并随机分组:生理盐水组、细辛低剂量组、细辛中剂量组、细辛高剂量组。分别在给药前、药物实验开始后3d、1w、2w、3w测量心率,并对比各组疗效。 结果:(1)氢氧化钠介入法制作的大鼠模型的病生理改变均符合病窦标准,且缩短了造模时间及减小了损伤;(2)细辛各剂量组心率改善情况如下:细辛高剂量组细辛中剂量组细辛低剂量组生理盐水组。 结论:(1)改进后的氢氧化钠介入法制作大鼠病态窦房结综合症模型方法简化了以往的造模方法,提高了模型的稳定性,降低了对模型的损伤程度,使建立模型的成功率有了进一步的提高;(2)细辛对改善大鼠病态窦房结综合症心率有一定疗效。
[Abstract]:Objective to develop a simple and accurate rat model of sick sinus syndrome (SSS), to further improve the method of modeling, simplify the experimental operation and increase the success rate of modeling. (2) to study the effect of Asarum on heart rate in sick sinus syndrome model. (3) to explore the mechanism of Asarum on the rat model of sick sinus node syndrome (SSS), and to analyze the mechanism of its effect on heart rate in sick sinus syndrome (SSS). Methods the healthy rats were screened by electrocardiogram, and 40 rats with HR of 350-450 beats / min were selected. After anesthesia with ether, the skin of the second intercostal region of the sternum of the rats was cut off about 1 mm. The self-made measuring electrode catheter was inserted through the second intercostal of the right margin of sternum close to the orifice, and the place of administration was determined according to the waveform of electrocardiogram (ECG). The rat model of sSS was established by injecting sodium hydroxide into the microinjector through self-made measuring electrode catheter. Finally, complete purse suture. (2) from 40 rat models of sick sinus syndrome, 32 successful model rats were selected and randomly divided into normal saline group, low dose group and middle dose group. The heart rate was measured 3 days before administration and 3 days after the start of the drug experiment in the high dose group. The heart rate was measured and the curative effect was compared in each group. Results (1) the pathological changes of the rat model made by sodium hydroxide interventional method were in accordance with the standard of sick sinus, and the time of modeling was shortened and the injury was reduced. (2) the heart rate of each dose group was improved as follows: high dose group, medium dose group, low dose group, normal saline group. Conclusion (1) the modified sodium hydroxide interventional method simplifies the previous modeling method, improves the stability of the model and reduces the degree of damage to the model. The success rate of modeling has been further improved; 2) Asarum has a certain effect on improving the heart rate of sick sinus node syndrome in rats.
