大鼠脑损伤后血浆ET-1的含量变化与损伤时间的关系

【摘要】:目的:应用酶联免疫法、放射免疫法定量测定SD大鼠脑损伤后血浆ET-1的表达规律。寻找ET-1的表达与损伤时间的关系,为法医学脑损伤的时间推断提供更为完善的方法。 方法:选取健康成年SD大鼠64只,雌雄不限,随机分为正常组(n=8),假损伤对照组(n=8)以及损伤实验组(n=48),实验组又按损伤时间不同分为6个亚组。实验组大鼠实施麻醉后,参照Feeney s法,采用自由落体打击装置,,于颅骨正中线旁3mm处造成脑损伤模型。于损伤后1h、6h、24h、48h、3d、7d六个时间点(n=8)采血4ml及2ml,离心分离血浆后置-20℃冰箱保存。正常组不做任何处理,假损伤对照组仅做钻孔处理,不做自由落体打击,不致脑损伤。应用酶联免疫法以及放射免疫法对大鼠脑损伤后血浆ET-1进行定量检测,并对检测结果进行统计学分析。 结果:(1)ELISA法:大鼠脑损伤后血浆ET-1含量于伤后1h开始升高(P0.05),与损伤后24h达到高峰。损伤后48h开始逐渐下降,3d后开始逐渐趋于假损伤组水平(P0.05),7d后基本恢复正常值。(2)RIA法:大鼠脑损伤后血浆ET-1含量表达规律基本与ELISA法所测结果表达规律一致。 结论:大鼠脑损伤后血浆ET-1的含量随损伤时间呈规律性变化。两种检测方法所得结果呈一致性。酶联免疫法及放射免疫法操作简单易行,可作为日后法医学脑损伤鉴定及研究的依据。
[Abstract]:Objective: to investigate the relationship between the expression of ET-1 and the time of brain injury in SD rats by enzyme linked immunosorbent assay (Elisa) and radioimmunoassay (RIA), and to provide a more perfect method for estimating the time of brain injury in forensic medicine. Methods: 64 healthy adult SD rats, male and female, were randomly divided into normal group (n = 8) and sham injury control group (n = 48). The experimental group was also divided into 6 subgroups according to the time of injury. The experimental group was anesthetized. According to Feeney s method, the model of brain injury was made at 3mm near the midline of skull by using free fall device. The blood samples were collected at 6 h, 6 h, 24 h, 48 h, 3 d, 7 d, six time points, 6 ml and 2 ml, then centrifuged plasma was stored in refrigerator at -20 鈩
