盐负荷和碱负荷对孕晚期大鼠离体子宫收缩力和催产素受体水平的影响

【摘要】:目的探讨盐负荷和碱负荷对孕晚期大鼠离体子宫收缩力和催产素受体(OXTR)水平的影响。方法将42只孕晚期大鼠随机均分为3组,A组饮用自来水,B组饮用2%Na Cl溶液,C组饮用5%Na HCO3溶液;分别于0、12、24、36、48、60和72 h时从3组中各随机选取2只大鼠麻醉,先剖腹采腹主动脉血行动脉血气分析,再将子宫纵向剖取数个均等肌条,检测其肌张力,最后采用ELISA法测定肌条中OXTR的含量。结果 3组在0 h时血Na+、HCO3-浓度差异均无统计学意义(P0.05);在其余各时间点,B组和C组血Na+浓度均显著高于A组(P0.05),C组血HCO3-浓度显著高于A组和B组(P0.05)。3组在0 h时子宫肌条收缩的振幅和频率差异均无统计学意义(P0.05);B组在其余各时间点的振幅和频率(除72 h振幅外)均显著高于A组(P0.05);C组在12、24和60 h时振幅显著高于A组(P0.05),在48、60和72 h时频率显著高于A组(P0.05)。3组0 h时OXTR含量差异均无统计学意义(P0.05);其余各时间点,B组OXTR含量显著高于A组(P0.05);C组在12、24、60和72 h时OXTR含量显著高于A组(P0.05)。结论盐负荷可使子宫收缩振幅、频率及子宫表面OXTR表达显著增强;碱负荷60 h时可使子宫收缩振幅、频率及OXTR表达三者均显著增强。
[Abstract]:Objective to investigate the effects of salt and alkali loading on uterine contractility and oxytocin receptor (oxytocin receptor) level in vitro of late pregnant rats.Methods 42 late pregnant rats were randomly divided into three groups: group A drinking tap water, group B drinking 2%Na Cl solution and group C drinking 5%Na HCO3 solution.The arterial blood gas of abdominal aorta was analyzed by laparotomy, and then several equal muscle strips were dissected longitudinally, and the muscle tension was detected. Finally, the content of OXTR in muscle strips was determined by ELISA method.Results at 0 h, there was no significant difference in serum Na ~ (2 +) -HCO ~ (3-) concentration among the three groups, and at other time points, the serum Na concentration of group B and group C was significantly higher than that of group A (P 0.05) and group B (P 0.05c) at 0 h, and that of group B was significantly higher than that of group A and group B (P 0.05.3) at 0 h.There was no significant difference in the amplitude and frequency of muscle strip contraction. The amplitude and frequency of group B (except 72 h amplitude) at other time points were significantly higher than those of group A at 1224 h and 60 h, respectively, and the amplitudes of group B were significantly higher than those of group A at 24 h and 60 h.The frequency at h was significantly higher than that in group A at 0 h, and the content of OXTR in group B was significantly higher than that in group A at 12: 24 and 72 h, and the content of OXTR in group B was significantly higher than that in group A at 0 h.Conclusion the amplitude and frequency of uterine contraction and the expression of OXTR on uterine surface were significantly increased by salt loading, and the amplitude, frequency and OXTR expression of uterine contraction were significantly increased at 60 h of alkali loading.
