粪肠球菌对大鼠根尖周炎NF-κB基因表达影响的研究

【摘要】：目的：通过建立大鼠根尖周炎模型，研究粪肠球菌对大鼠根尖周炎NF-κB p65表达的影响。方法：选取健康雄性SD大鼠24只，使用小号球钻于右侧下颌第一磨牙近中窝行开髓术后，随机分为A、B两组，每组各12只。A组开髓后，髓腔内封入PBS缓冲液小棉球，设为未封菌组；B组开髓后，髓腔内封入粪肠球菌与PBS缓冲液调制的菌悬液小棉球，设为封菌组。两组均使用进口玻璃离子粘固剂封闭髓腔。左侧下颌第一磨牙为空白对照组。常规条件饲养，A、B两组分别于术后1周、2周、3周、4周四个时间点随机各取3只鼠，水合氯醛麻醉处死后，分离其下颌骨，用小号挖匙挖取右侧下颌第一磨牙和左侧下颌第一磨牙根尖区牙槽骨、牙周膜组织，之后对组织进行研磨，，提取、纯化组织RNA，并逆转录成cDNA，采用实时荧光定量PCR (Real-time PCR)手段检测各组NF-κB p65基因的表达水平并进行统计学分析。结果： 1.健康空白对照组的NF-κB p65mRNA有少量表达，1周到4周A、B两组各时间点的NF-κB p65mRNA的表达量均高于相应空白对照组，差异有统计学意义。（P0.05） 2．A、B两组术后1周到2周NF-κB p65mRNA的表达水平逐渐增高，2周表达水平达最高峰，3周表达水平开始降低，4周表达水平继续降低。四个实验组中1周与4周之间的差异无统计学意义（P0.05），其余各组之间的差异均具有统计学意义（P0.05）。 3．术后2周、3周，B组NF-κB p65mRNA的表达量均高于A组，差异均具有统计学意义（P0.05）。结论： 1．NF-κB p65基因的表达水平可能与根尖周炎的发生、发展密切相关，提示NF-κB p65参与了根尖周炎的炎症反应和骨破坏。 2．NF-κB p65与粪肠球菌感染为主的根尖周炎密切相关，提示粪肠球菌存可能加重了根尖周组织的炎症反应和骨破坏。
[Abstract]:Aim: to study the effect of Enterococcus faecalis on the expression of NF- 魏 B p65 in rat periapical periodontitis. Methods: Twenty-four male Sprague-Dawley (SD) rats were randomly divided into two groups: group A (n = 12) and group A (n = 12). PBS buffer was injected into the medullary cavity of 24 male Sprague-Dawley rats. After opening the pulp in group B of unsealed bacteria, the bacteria suspension of Enterococcus faecalis and PBS buffer was added into the medullary cavity. Both groups were treated with imported glass ionomer to seal the medullary cavity. The left mandibular first molar was a blank control group. Three rats were randomly selected from each group at the time points of 1 week, 2 weeks, 3 weeks and 4 days after routine feeding. The mandible was isolated after anesthesia with chloral hydrate. The alveolar bone and periodontal ligament of the right first molar and the left first molar of the right mandible were extracted with a small spoon, and then the tissue was ground and extracted. The expression level of NF- 魏 B p65 gene in each group was detected by real-time fluorescence quantitative PCR real-time PCR, and the expression level of NF- 魏 B p65 gene was analyzed statistically. Results: 1. The expression of NF- 魏 B p65mRNA in the healthy blank control group was significantly higher than that in the control group at each time point from 1 to 4 weeks. The difference was statistically significant (P 0.05). 2. The expression level of NF- 魏 B p65mRNA increased gradually from 1 week to 2 weeks after operation in both groups. The expression level of NF- 魏 B increased gradually at the end of 2 weeks. The expression level of NF- 魏 B began to decrease at the end of 3 weeks and the expression level of NF- 魏 B decreased at the 4th week. There was no significant difference between one week and four weeks in the four experimental groups, but the differences among the other groups were statistically significant. 3. The expression of NF- 魏 B p65mRNA in group B was significantly higher than that in group A at 2 weeks and 3 weeks after operation, and the difference was statistically significant (P 0.05). Conclusion: 1. The expression level of NF- 魏 B p65 gene may be closely related to the occurrence and development of periapical inflammation, suggesting that NF- 魏 B p65 is involved in the inflammatory response and bone destruction in periapical periodontitis. 2. NF- 魏 B p65 was closely related to periapical periodontitis with Enterococcus faecalis infection, suggesting that the presence of Enterococcus faecalis may aggravate inflammation and bone destruction in periapical tissue.
