当归挥发油对大鼠胃肠平滑肌肌电活动的影响

【摘要】:目的: 本实验旨在研究观察当归挥发油对大鼠胃窦及十二指肠平滑肌肌电活动的影响,分析当归挥发油对胃肠平滑肌的作用特点及规律,为探讨当归挥发油在防治消化道疾病方面的价值提供参考。 方法: Wistar大鼠禁食不禁水18h后,10%水合氯醛麻醉,手术暴露胃及小肠,分别用银针电极自胃窦及十二指肠垂直于纵行肌斜刺入浆膜下2mm肌层,然后与BL-420F生物机能实验系统相连接,建立胃肠平滑肌肌电实验研究系统。设立阿托品组、维拉帕米组、多巴胺组为工具药,以胃窦及十二指肠平滑肌慢波肌电的频率和振幅变化等为观察指标,观察当归挥发油对正常麻醉大鼠在体胃窦及十二指肠平滑肌慢波肌电活动、对新斯的明诱导的大鼠胃窦及十二指肠平滑肌慢波肌电活动、对氯化钡诱导大鼠胃窦及十二指肠平滑肌慢波肌电活动及对吗丁啉诱导的大鼠胃窦及十二指肠平滑肌慢波肌电活动的影响。 结果: 1)当归挥发油中剂量(0.2%)和低剂量(0.1%)下,麻醉大鼠胃窦平滑肌慢波肌电频率和振幅呈现抑制效应,而十二指肠平滑肌慢波肌电的频率和振幅呈现兴奋效应;当归挥发油高剂量(0.4%)胃窦平滑肌慢波肌电的频率和振幅呈现兴奋效应,,而十二指肠平滑肌慢波肌电的频率和振幅则呈现抑制效应。2)当归挥发油0.4%、0.2%、0.1%三个剂量组对新斯的明诱导的麻醉大鼠胃窦和十二指肠平滑肌慢波肌电频率和振幅的变化均表现为明显的拮抗作用,其作用曲线与拮抗药阿托品的作用曲线相一致。3)当归挥发油0.2%、0.14%、0.1%三个剂量组对氯化钡诱导的十二指肠平滑肌慢波肌电频率的变化呈现明显的拮抗作用,且效应曲线特征与维拉帕米表现为高度的一致性。4)当归挥发油高剂量组对吗丁啉诱导的麻醉大鼠胃窦和十二指肠平滑肌慢波肌电频率的变化呈现明显的拮抗作用,且效应曲线特征与多巴胺表现为高度的一致性。 结论: 1)当归挥发油对麻醉大鼠胃窦和十二指肠平滑肌正常慢波肌电频率和振幅具有相反的作用;2)不同浓度当归挥发油对麻醉大鼠胃窦和十二指肠平滑肌正常慢波肌电频率和振幅具有“双向”作用;3)当归挥发油对新斯的明诱导的麻醉大鼠胃窦和十二指肠平滑肌肌电慢波频率和振幅的变化呈现明显拮抗作用,该作用可能与阻滞M受体有关;4)当归挥发油对BaCl2诱导的麻醉大鼠十二指肠平滑肌慢波肌电频率的变化呈现明显的拮抗作用,该作用可能与阻滞钙离子通道有关。5)当归挥发油对吗丁啉诱导的麻醉大鼠十二指肠平滑肌慢波肌电频率的变化呈现明显的拮抗作用,该作用可能与兴奋多巴胺受体有关。
[Abstract]:Objective: The purpose of this study was to observe the effect of volatile oil of Angelica sinensis on the myoelectric activity of gastric antrum and duodenal smooth muscle in rats, and to analyze the characteristics and regularity of the effect of volatile oil of Angelica sinensis on gastrointestinal smooth muscle. To explore the value of essential oil of Angelica sinensis in the prevention and treatment of digestive tract diseases. Methods: After fasting for 18 hours, Wistar rats were anesthetized with 10% chloral hydrate. The stomach and small intestine were exposed. Silver needle electrodes perpendicular to the longitudinal muscle from antrum and duodenum were applied to the subserous 2mm muscle layer, and then connected with the BL-420F biological function experimental system. To establish an experimental study system of gastrointestinal smooth muscle electromyography. Atropine group, verapamil group and dopamine group were used as tool drugs. The changes of frequency and amplitude of slow wave myoelectric activity in gastric antrum and duodenal smooth muscle were observed. To observe the slow wave myoelectric activity of gastric antrum and duodenal smooth muscle in normal anesthetized rats, and to observe the slow wave myoelectric activity of gastric antrum and duodenal smooth muscle induced by neostigmine. The effects of barium chloride on the slow wave myoelectric activity of gastric antrum and duodenal smooth muscle and morpholine induced slow wave myoelectric activity of gastric antrum and duodenal smooth muscle of rats were studied. Results: 1) the frequency and amplitude of slow wave myoelectricity in antral smooth muscle of anesthetized rats showed inhibitory effect, but the frequency and amplitude of slow wave electromyography in duodenal smooth muscle showed excitatory effect. The frequency and amplitude of slow wave electromyography of gastric antral smooth muscle showed excitatory effect. The frequency and amplitude of slow wave myoelectric activity in duodenal smooth muscle showed inhibitory effect. 2) the changes of frequency and amplitude of slow wave myoelectric activity induced by neostigmine in antral and duodenal smooth muscle of anesthetized rats treated with 0. 4% volatile oil of Angelica sinensis and 0. 1% 0. 1% of Angelica sinensis All of them showed obvious antagonistic effects. The action curve was consistent with that of antagonist atropine. 3) the volatile oil of Angelica sinensis 0.20.14and 0.1% had obvious antagonistic effect on the changes of slow wave frequency of duodenal smooth muscle induced by barium chloride. The characteristics of the effect curve were highly consistent with verapamil. 4) the high dose group of Angelica sinensis volatile oil showed obvious antagonistic effect on the changes of slow wave electromyography frequency of gastric antrum and duodenum smooth muscle induced by morpholine in anesthetized rats. The characteristics of the response curve were highly consistent with that of dopamine. Conclusion: 1) the volatile oil of Angelica sinensis has the opposite effect on the frequency and amplitude of normal slow wave myography in antrum and duodenal smooth muscle of anesthetized rats. (2) different concentrations of volatile oil of Angelica sinensis on normal slow wave myoelectric activity of gastric antrum and duodenal smooth muscle of anesthetized rats The frequency and amplitude have a "bidirectional" effect. The volatile oil of Angelica sinensis has a significant antagonistic effect on the changes of frequency and amplitude of myoelectric slow wave of gastric antrum and duodenal smooth muscle induced by neostigmine in anesthetized rats. This action may be related to blocking M receptor. The volatile oil of Angelica sinensis has an obvious antagonistic effect on the frequency of slow wave electromyography of duodenal smooth muscle induced by BaCl2 in anesthetized rats. This action may be related to blocking calcium channel. 5) Angelica volatile oil has obvious antagonistic effect on the changes of slow wave electromyography frequency of duodenal smooth muscle induced by morpholine in anesthetized rats, which may be related to excitatory dopamine receptor.
